The effect of four stereoisomers of steroidal amidoesters with p-N, N-bis (2-chloroethyl)aminophenylbutyric acid (Chlorambucil) has been tested on sister chromatid exchanges (SCE) and on human lymphocyte proliferation kinetics, A correlation was observed in this group of four compounds between the magnitude of the SCE response and the depression in the proliferation rate index, The antitumour activity of the same compounds was tested on leukemia P388-bearing mice and on DNA synthesis of P388 and L1210 leukemia cell cultures, We observed that the order of the antitumour effectiveness of the four-compounds tested coincided with the order of the cytogenetic effects they induced.
