Στο πρώτο μέρος αυτής της διατριβής περιγράφεται η παρασκευή μιας σειράς νανοϋβριδικών υλικών με χαρακτηριστικές μαγνητικές, χημικές και βιολογικές ιδιότητες. Η σχεδίαση των υλικών αυτών είχε σαν απώτερο σκοπό την χρησιμοποίηση τους σε βιοιατρικές εφαρμογές και ειδικότερα στην απεικόνιση μαγνητικού συντονισμού (MRI) σαν σκιαγραφικά μέσα και τη θεραπεία σύλληψης νετρονίων (NCT). Σαν αρχικό υλικό επιλέχθηκαν τα στοιβαγμένα διπλά υδροξείδια (LDH) και συντέθηκε μια ποικιλία από διαφορετικά μέλη της οικογένειας αυτής. Ιδιαίτερο βάρος δόθηκε στην εισαγωγή παραμαγνητικών ιόντων στο σκελετό των ανόργανων σωματιδίων, όπως ιόντα του μαγγανίου και του σιδήρου. Η προσπάθεια σύνθεσης LDH με στοιχεία της ομάδας των λανθανίδων αποτέλεσε κομβικό σημείο στην ανάπτυξη δύο εντελώς ανόμοιων υλικών με εξαιρετικές ιδιότητες, των Gd2(OH)5NO3 και Gd(OH)3. Το Gd2(OH)5NO3 ανήκει στην οικογένεια των υδροξυνιτρικών, μια εντελώς νέα κατηγορία στοιβαγμένων υδροξειδίων με φυλλόμορφη δομή και ανιοανταλλακτικές ιδιότητες. Το Gd2(OH)5NO3 παρουσιάζει όμοια δομικά χαρακτηριστικά με αυτά του LDH. Tο Gd(OH)3 αποτελείται από δέσμες παράλληλων νανοράβδων και βρίσκει πολυάριθμες εφαρμογές στην επιστήμη και την τεχνολογία, λόγω των ιδιαίτερων οπτικών, ηλεκτρικών και χημικών ιδιοτήτων τους. Η δομή του Gd(OH)3 είναι εξαγωνική P63/m και τα άτομα του γαδολινίου στοιχίζονται σε παράλληλες γραμμικές αλυσίδες. Η επιφάνεια του Gd2(OH)5NO3 τροποποιήθηκε έτσι ώστε να αποκτήσει υδατοδιαλυτότητα και παράλληλα στόχευση σε καρκινικά κύτταρα. Αυτό επιτεύχθηκε με τη δέσμευση μορίων φολικού οξέος. Περαιτέρω τροποποίηση, με ομοιοπολική δέσμευση μορίων όπως της βιοτίνης και του ATP στην επιφάνεια του σωματιδίου, επιτρέπει στο νανοϋβρίδιο να βρίσκει σημαντικές εφαρμογές. Επιπροσθέτως, στο χώρο μεταξύ των στοιβάδων του υλικού εισήχθησαν μόρια όπως το αντιβιοτικό ναλιδιξικό οξυ, τα αμινοξέα ασπαρτικό και γλουταμικό και το λιπαρό οξύ, παλμιτικό. Η τροποποίηση αυτή επιτρέπει την εφαρμογή των νανοϋβρίδιων σε συστήματα μεταφοράς φαρμάκων. Το Gd(OH)3 τροποποιήθηκε έτσι ώστε να αποκτήσει βιοσυμβατότητα. Αυτό έγινε δυνατό με επικάλυψη της επιφάνειας του με πολύ-λακτικό οξύ (PLA). Μια δεύτερη τροποποίηση της επιφάνειας του Gd(OH)3 πραγματοποιήθηκε έπειτα από ομοιοπολική δέσμευση του αμινοξέος τρυπτοφάνη. Τα σωματίδια αυτά δεν παρουσιάζουν τοξικότητα έναντι του βακτηρίου E.coli και του μύκητα Sacharomyces Cerivisae. Παράλληλα διέθεταν την ικανότητα να αυξάνουν τον ρυθμό χαλάρωσης Τ1 των πρωτονίων του H2O. Τέλος, τα νανοϋβρίδια χαρακτηρίστηκαν με περίθλαση ακτινών Χ, φασματοσκοπία υπερύθρου, φασματοσκοπία υπεριώδους-ορατού, φασματοσκοπία EDS, μικροσκοπία TEM, HRTEM και SEM καθώς επίσης με θερμοσταθμική ανάλυση. Το δεύτερο μέρος αναφέρεται στη σύνθεση ενός καλά χαρακτηρισμένου υλικού, της silica τύπου MCM-41. Ακολούθησε, η ακινητοποίηση του ενζύμου οξειδάσης της γλυκόζης (GOx) στη silica. Το νανοϋβρίδιο χαρακτηρίστηκε με περίθλαση ακτινών Χ, φασματοσκοπία υπερύθρου, μικροσκοπία TEM και θερμοσταθμική ανάλυση. Τέλος, προσδιορίστηκαν οι κινητικοί παράμετροι του ακινητοποιημένου και ελεύθερου ενζύμου και παρασκευάστηκε ένας βιοαισθητήρας του ακινητοποιημένου ενζύμου. (EL)
In the first part of the particular study, a series of nanohybrid materials was prepared, which has present characteristic magnetic, chemical and biological properties. These nanohybrids were appropriately designed for biomedical applications such as magnetic resonance imaging (MRI) and neutron capture therapy (NCT). Double layered hydroxides (LDH) were the base of our synthesis, preparing a variety of different types, after the introduction of paramagnetic elements in the inorganic framework. The change-overs of lanthanide ions allow the preparation of two completely disparate materials, Gd2(OH)5NO3 and Gd(OH)3. Gd2(OH)5NO3 consist of a new class of anion exchangeable layered hydroxides incorporating gadolinium cations within the layers. This material has the potential to combine the properties of the lanthanides with the flexibility of intercalation hosts. The surface of Gd2(OH)5NO3 was modified in order to obtain water solubility, functionalization and the ability to target cancer cells. This was achieved by covalent bonding of folic acid or biotin on the surface of Gd2(OH)5NO3. In addition, a variety of molecules, were intercalated in Gd2(OH)5NO3, such as the antibiotic nalidic acid, the aminoacids aspartate and glutamate as well as the fatty acid palmitic acid. Gd(OH)3 was prepared during a variation of the above synthesis. Gd(OH)3 is consisted of bundle-like nanorods and has numerous applications in science and technology, due to its special optical, electrical and chemical properties. In order to produce a biocompatible material, a poly-lactate layer was developed. Furthermore, the aminoacid tryptophan was covalent bonded on the surface of the particles. Experiments show that these materials did not present toxicity towards to the bacterium E.coli and the Sacharomyces Cerivisae. Apart from that they present a meaningful increase of proton relaxivity and are considered to be promising particles for proportional purposes. The above nanohybrids were characterized by using X-ray diffraction, infrared spectroscopy, ultraviolet-visible spectroscopy, magnetic measurements, TEM, HRTEM and SEM microscopy as well as thermogravimetric analysis. In the second part of this study, is mentioned the immobilization of GOx on the surface of silica MCM-41 and the extensive characterization of the immobilized enzymes. The nanohybrid was characterized by using X-ray diffraction, infrared spectroscopy, TEM microscopy as well as thermogravimetric analysis.
