Η γνωστική ανάπτυξη του νεογνού καθορίζεται από την αλληλεπίδρασή του με τη
μητέρα του. Χρησιμοποιώντας τη μητρική επαφή ως θετικό ή ως αρνητικό ενισχυτή,
νεογνά επίμυος, στα οποία είτε αποτράπηκε η επαφή με τη μητέρα (ομάδα
συνεχόμενης ματαίωσης), είτε δόθηκε ως ανταμοιβή (ομάδα συνεχόμενης ενίσχυσης),
βελτίωσαν σταδιακά τις επιδόσεις του, μετά από εκπαίδευση τεσσάρων ημερών
(μεταγεννητικές ημέρες 10-13, Ρ10-13) σε ένα λαβύρινθο σχήματος Τ. Μάλιστα, τα
«επιβραβευμένα» νεογνά εντόπιζαν τη μητέρα τους ταχύτερα από τα «ματαιωμένα».
Στην παρούσα εργασία διερευνήθηκε η υπόθεση της πλαγίωσης και η επίπτωση των
δύο παραπάνω πρώιμων εμπειριών σε σχέση με ζώα μάρτυρες, στο πρωτέωμα του
προμετωπιαίου φλοιού, μια περιοχή που έχει δειχθεί ότι επηρεάζεται από αυτές
τις συγκεκριμένες πρώιμες εμπειρίες και είναι πολύ σημαντική για την μνήμη
εργασίας και τη συμπεριφορά.
Συνολικά, ταυτοποιήθηκαν 117 πρωτεΐνες. Η πλειονότητα των πρωτεϊνών,
συγκεκριμένα 72 από τις 117 ταυτοποιημένες πρωτεΐνες, βρέθηκε να προέρχεται από
το κυτταρόπλασμα. Όσον αφορά τις κυτταροσκελετικές πρωτεΐνες βρέθηκαν
αξιοσημείωτες διαφορές, συγχρόνως ένας αριθμός πρωτεϊνών που εμπλέκονται στην
κυτταρική μορφογένεση, νευρογένεση και την απόπτωση βρέθηκε ότι εκφράζεται
διαφορετικά μεταξύ των τριών κατηγοριών επίμυων που μελετήθηκαν. Επίσης, ένα
μεγάλο μέρος των αλλαγών που παρατηρήθηκαν ως αποτέλεσμα των πρώιμων εμπειριών
στη μελέτη μας σημειώθηκε σε πρωτεΐνες διάμεσου μεταβολισμού.
Στο επίπεδο μελέτης δεξιάς και αριστερής πλαγίωσης που έγινε στην παρούσα
μελέτη βρέθηκε ότι στα ζώα μάρτυρες παρατηρήθηκε υπερέκφραση πρωτεϊνών στον
δεξιό σε σχέση με τον αριστερό προμετωπιαίο φλοιό, στα επιβραβευμένα δεν
παρατηρήθηκε σημαντική διαφορά στον αριθμό υπερεκφραζόμενων πρωτεϊνών μεταξύ
των 2 ημισφαιρίων, ενώ στα ματαιωμένα διαπιστώθηκε υπερέκφραση πρωτεϊνών στον
αριστερό σε σχέση με τον δεξιό προμετωπιαίο φλοιό.
Τα ευρήματα της παρούσας μελέτης προτείνουν ότι οι αλλαγές στη νευρωνική
πλαστικότητα που προκαλούνται από τις πρώιμες εμπειρίες μπορεί να θεωρηθούν ως
το τελικό αποτέλεσμα ενεργοποίησης και/ή αναστολής μοριακών καταρρακτών όπως
αυτοί που ρυθμίζουν τη δυναμική του κυτταροσκελετού, την ενδοκυττάρια μεταγωγή
σήματος και το διάμεσο μεταβολισμό.

During the neonatal period of rodents, mother-infant interaction mediates
environmental stimulation of the pups and cognitive and emotional development.
When mother contact is used as either a positive or a negative reinforcement,
rat pups, both those denied contact (frustrated) and those allowed contact as
continuous reward (rewarded), improve their performance in a T maze after four
days of training (postnatal day 10-13). Rewarded pups however, are able to
locate their mother faster than frustrated ones.
In this research we investigate the hypothesis of laterality and the effect of
these early experiences on the proteome of the prefrontal cortex, an area
crucial for working memory and behavior, which is affected in our model as
shown by previous results and. We also investigate and explore the expression
of proteins in both the right and left hemispheres of the prefrontal cortex in
each group separately (control, rewarded and frustrated) by using a high
performance technique: Combined results of two-dimensional electrophoresis and
mass spectrometry are used for proteomic analysis.
Using this analysis, a total of 117 proteins, affected by early experiences,
were identified. The majority of the proteins, specifically 72 out of 117,
originated from the cytoplasm. Regarding the cytoskeletal proteins,
significant differences were found between the three groups in terms of the
expression of actin and tubulin isoforms. Significant differences were also
observed in the left prefrontal cortex in the expression of alpha-internexin,
of the glial fibrillary acidic protein (GFAP) and of the
dihydropyrimidinase-related protein 2 (DRP-2). A number of proteins involved
in cell morphogenesis, neurogenesis and apoptosis, such as 14-3-3 proteins, and
several heat shock proteins (HSP) were found to be differently expressed among
the three groups studied. Also, a large number of changes in intermediate
metabolism proteins, such as the enzyme complex of adenosine triphosphate
synthase, the glyceraldehyde 3-phosphate dehydrogenase (GAPDH), the
phosphoglycerate mutase 1 and the triosephosphate isomerase were observed.
Regarding laterality an overexpression of proteins in the right hemisphere of
the prefrontal cortex of control animals was observed. No significant
difference was observed in the number of overexpressed proteins in the two
hemispheres in the rewarded rat pups. In contrast, in frustrated pups there
was an increase in the number of overexpressed proteins in the left hemisphere
of their prefrontal cortex.
Differential and asymmetric over-expression in the two hemispheres of the
prefrontal cortex of proteins from different functional categories suggests
that laterality may be a universal phenomenon in this brain region during early
development. The results from our study indicate a significant influence of
early experiences on brain laterality and on a series of paths, such as energy
metabolism, the mechanism of cytoskeletal organization and neurogenic and
apoptotic mechanisms.
Our findings suggest that changes in neuronal plasticity induced by early
experiences may represent the final result of activation and/or supresion of
molecular cascades involved in the regulation of cytoskeletal dynamics,
intracellular signal transduction as well as of intermediate metabolism.
