The effects of gestazional and lactational choline derivation on brain function of the offspring rats
Επιπτώσεις δίαιτας ανεπαρκούς σε χολίνη κατά την κύηση σε εγκεφαλικές λειτουργίες απόγονων επιμύων

Σκοπός: Το θέμα της μελέτης είναι η διερεύνηση επίδρασης της διαιτητικής ανεπάρκειας χολίνης (choline deficient diet, CDD) σε συγκεκριμένα σημαντικά ένζυμα (AChE, Να⁺ -Κ⁺ ΑΤΡάση Mg²⁺ ΑΤΡάση) και της ολικής αντιοξειδωτικής ικανότητας (total antioxidant status, TAS), σε ομογενοποίημα εγκεφάλου απογόνων επιμύων (νεογέννητων, 21 ημερών και ενήλικων) που έλαβαν δίαιτα CDD. Επιπλέον μελετήθηκε ο ρόλος της ομοκυστεΐνης και 2 αμινοξέων της L-φαινυλαλανίνης και L-αλανίνης. Καθώς η μνήμη και η συμπεριφορά επηρεάζονται από την ACh, μελετήθηκε η επίδραση της δίαιτας CDD στη γνωσιακή συμπεριφορά των πειραματόζωων με τη δοκιμασία αναγνώρισης αντικειμένων, ORT. Πειραματόζωα-Μεθοδολογία: Χρησιμοποιήθηκαν θηλυκοί επίμυες ράτσας Albino Wistar rats βάρους 170±25 g. Μετά την σύζευξη οι θηλυκοί επίμυες τοποθετήθηκαν σε ξεχωριστούς κλωβούς και ταξινομήθηκαν σύμφωνα με τις ομάδες των πειραματικών πρωτοκόλλων σε: C=ομάδα μαρτύρων υπό ισορροπημένη δίαιτα (ND), (Ν=9), και CD=ομάδα μελέτης υπό δίαιτα ανεπαρκή σε χολίνη CDD (Ν=9). Προετοιμασία ιστού εγκεφάλου: Τα πειραματόζωα θανατώθηκαν με ακαριαίο αποκεφαλισμό και οι εγκέφαλοι απομακρύνθηκαν γρήγορα. Μετά την προετοιμασία του ιστού σύμφωνα με (Tsakiris 2001, Tsakiris et al 2000), μετρήθηκαν οι ενζυμικές δραστικότητες των: AChE (Ellman et al. 1961, Tsakiris, 2001), Na⁺, K⁺-ΑΤΡάσης and Mg²⁺-ATPάσης (Bowler and Tirri 1974; Tsakiris 2001) και του TAS (Carageorgiou et al. 2003, 2004). H μελέτη περιελάμβανε 3 πειραματικά πρωτόκολλα, Α, Β, Γ. Πειραματικό πρωτόκολλο Α: Έγιναν 3 διαφορετικά επιμέρους πειράματα Ι, II, III. Πείραμα Ι: ομάδα CD: χορηγήθηκε δίαιτα CDD κατά την κύηση. ομάδα C: χορηγήθηκε δίαιτα ND κατά την κύηση. Πείραμα II: ομάδα CD: χορηγήθηκε η δίαιτα CDD κατά την κύηση και η ισορροπημένη δίαιτα (ND) κατά τον θηλασμό. ομάδα C: χορηγήθηκε δίαιτα ND. Πείραμα III: ομάδα CD: χορηγήθηκε η δίαιτα CDD κατά την περίοδο της κύησης και του θηλασμού (Συνολική διάρκεια 6 εβδομάδων). ομάδα C: χορηγήθηκε δίαιτα ND. Στο κάθε πείραμα τα αποτελέσματα των ομάδων CD συγκρίθηκαν με την ομάδα C στην οποία χορηγήθηκε ισορροπημένη δίαιτα (ND). Πειραματικό πρωτόκολλο Β: Περιελάμβανε 2 διαφορετικά επιμέρους πειράματα Ι, II, και το in vitro πείραμα III. -Πείραμα Ι ομάδα CD: χορηγήθηκε δίαιτα CDD κατά την κύηση\και θηλασμό καθώς και για 3 εβδομάδες μετά το τέλος του. ομάδα C: χορηγήθηκε δίαιτα ND. -Πείραμα II: ομάδα CD: χορηγήθηκε δίαιτα CDD κατά την κύηση και θηλασμό καθώς και για 3 εβδομάδες μετά το τέλος του θηλασμού, ενώ ακολούθησε χορήγηση ισορροπημένης δίαιτας για επιπλέον 18 εβδομάδες (θανάτωση επιμύων σε ηλικία 24 εβδομάδων). ομάδα C: χορηγήθηκε δίαιτα ND. Στο κάθε πείραμα τα αποτελέσματα των ομάδων CD συγκρίθηκαν με την ομάδα C στην οποία χορηγήθηκε ισορροπημένη δίαιτα (ND). Πείραμα III in vitro πειράματα από τους εγκεφάλους των άρρενων επίμυων από το πείραμα Ι. Πειραματικό πρωτόκολλο Γ: δοκιμασία αναγνώρισης αντικειμένων ORT. Στατιστική ανάλυση: χρησιμοποιήθηκε το t test και ANOVA. (στατιστικώς σημαντικό p<0.05). Αποτελέσματα: Πειραματικό πρωτόκολλο Α : παρατηρήθηκε μια στατιστικά σημαντική μείωση της δραστικότητας των ενζύμων AChE, Να⁺ -Κ⁺ ΑΤΡάσης 23%, 24% και 50%, αντίστοιχα και μείωση του TAS (-27%, p<0.001) ενώ δεν παρατηρήθηκε καμία διαφορά για την Μg²⁺ ΑΤΡάση.. Κατά την διάρκεια του θηλασμού παρατηρήσαμε επαναφορά της δραστικότητας των ενζύμων ενώ δεν παρατηρήθηκε το ίδιο για το TAS. Πειραματικό πρωτόκολλο Β: παρατηρήθηκε αύξηση της δραστικότητας των AChE, (+20%, p<0.001) και Να⁺ -Κ⁺ ΑΤΡάσης (+35%, p<0.001) ενώ παρατηρήθηκε μείωση της TAS (-29%, p<0.001) για τα πειραματόζωα ηλικίας 6 εβδομάδων. Μετά την αναστροφή της δίαιτας σε ισορροπημένη για το χρονικό διάστημα 18 εβδομάδων παρατηρήσαμε αποκατάσταση της δραστικότητας των παραπάνω ενζύμων καθώς και της TAS. Στα In vitro παρατηρήσαμε τον προστατευτικό ρόλο των δύο αμινοξέων L-αλανίνης και L-φαινυλαλανίνης καθώς και τη δράση της ομοκυστεḯνης (πιθανός αιτιολογικός παράγοντας των μεταβολών που παρατηρήθηκαν από την δίαιτα CDD). Συμπεράσματα: Η μελέτη μας πιθανολογεί ότι η μείωση της δραστικότητας των παραπάνω ενζύμων αποκαταστάθηκε μετά από 21 ημέρες (περίοδο θηλασμού) λόγω νέων μορφών συναπτογένεσης, και/ή ουσιών που λαμβάνονται κατά τον θηλασμό. Η χρόνια χορήγηση δίαιτας CDD κατά την κύηση, τον θηλασμό και επιπλέον 3 εβδομάδες προκάλεσε την αύξηση της ενζυμικής δραστικότητας των παραπάνω ενζύμων και μείωση του TAS, παράμετροι οι οποίοι αποκαταστήθηκαν μετά την αναστροφή της δίαιτας σε ισορροπημένη (για 18 εβδομάδες). Τα in vitro πειράματα η ομοκυστεḯνη φαίνεται να δίνει αποτελέσματα όμοια με εκείνα της χρόνιας πρόσληψης CDD, ενώ η L-αλανίνη και η L-φαινυλαλανίνης δρουν προστατευτικά στις αλλοιώσεις προκαλούμενες από την ομοκυστεΐνη, (ένδειξη του πιθανού προστατευτικού ρόλου των 2 αυτών αμινοξέων στις αλλοιώσεις προκαλούμενες από δίαιτα CDD). Η μείωση του χρόνου εξερεύνησης του νέου αντικειμένου της ομάδας CD σε σχέση με την ομάδα μαρτύρων, είναι ενδεικτική του σημαντικού ρόλου της χολίνης από τις πρώτες εμβρυḯκές μέρες, για την ανάπτυξη του εγκεφάλου.
The aim of the present study was to investigate how CDD administration during pregnancy and lactation could affect: a) total antioxidant status (TAS) and activities of AChE. (Na⁺, K⁺)-ATPase and Mg²⁺-ATPase on the brains of both male and female newborn and suckling (21-one day old) rats and (b) the above mentioned enzymes on the brains of older rats after 3 additional weeks of CDD and for an additional observation period of 18 weeks under equilibrated diet, moreover, the role of Hcy, and two other important amino-acids [L-phenylalanine (Phe) and L-alanine (Ala), both known to significantly affect the examined enzymatic parameters] (Schulpis et al 2006a, Tsakiris 2001) in certain of the above effects was examined, by undertaking in vitro experiments. Since memory is directly affected by Ach the effect of CDD on the memory of the rats exposed to CDD during pregnancy and lactation was examined by use of the Object Recognition Test (ORT) test. Animals and Methods: Animals: Albino Wistar rats weighing 170 ±25 g (purchased from the National Center of Scientific Research "Demokritos") were used in all experiments. Mating was performed between the purchased male and female rats. After mating, the female rats were placed in individual cages and were separated according to the protocol of each experimental procedure 1, 2, 3 aiming to investigate the effect of CDD on younger (1) and older rats (2) and the effect of CDD on memory (3). Expenmental procedure 1: Effect of CDD on younger rats. The protocol followed consisted of three experiments (Ι, ΙΙ, III). In experiment I, CDD was administered throughout gestation and brains were obtained from newborn offspring rats (of both sexes) immediately after delivery. In experiment II, CDD was administered throughout gestation and a normal diet was provided throughout lactation, brains were obtained from 21-day-old rats of both sexes. In experiment III, CDD was administered throughout gestation and lactation, while brains were also obtained from 21-day-old rats of both sexes. The CD group parameters were compared to their matched controls. Thus, for each experiment, four groups of rats (n=8 each) were used male controls (MC), female controls (FC), male choline-deprived (MCD) and female choline-deprived (FCD) rats. Experimental procedure 2. Effect of CDD on older rats: (a) group C (control group, n=9) that was fed with equilibrated diet, and (b) group CD (choline-deprived group, n=9) that was fed with CDD. The protocol followed consisted of two experiments I and II. In experiment I, CDD was administered throughout gestation and lactation, as well as for three more weeks (postlactationally); brains were obtained from 6-weeks-old offspring rats of both sexes. In experiment II, CDD was administered to the offspring rats throughout the same period as in experiment I, but equilibrated diet was administered for 18 additional weeks. In experiment II, brain tissues were obtained from 24-weeks-old offspring rats of both sexes. For each experiment, four groups of rats (n=8 each) were used to estimate the biochemical parameters: male controls (MC), female controls (FC), male choline deprived (MCD) and female choline deprived (FCD) rats. In vitro experiments were also performed in brain tissues acquired from experiment I male rats (MC and MCD rats). Exp Procedure 3: ORT (Object Recognition Test). The ORT was carried out according to the procedure described by Ennancer and Delacour. Four groups of rats (n=8) were used male controls (MC), female controls (FC), male choline deprived (MCD) and female choline deprived (FCD) rats. CDD was administered throughout gestation and lactation, as well as for three more weeks (postlactationally). Tissue preparation: The animals were sacrificed by decapitation and whole brains were rapidly removed. After tissue preparation (Tsakiris 2001; Tsakiris et al. 2000). The enzyme activities, namely AChE (Ellman et al. 1961, Tsakiris, 2001), Na⁺, K⁺-ATPase and Mg²⁺- ATPace activities (Bowler and Tirri 1974; Tsakiris 2001). and TAS (Carageorgiou et al. 2003, 2004) were measured. Statistical analysis: Data were analyzed using a paired t test and ANOVA. Statistical significance was considered for p values of <0.05. Results: Exp Procedure 1. In choline-deprived (CD) newborn rats, TAS and AChE and Na⁺, K⁺-ATPase activities were significantly reduced by 23%, 24% and 50%, respectively, in the brains of both sexes. Gestational CDD caused only a decrease in TAS (-27%, p<0.001) in suckling rat brains in both sexes. No changes were observed for the other enzyme activities. Moreover, gestational and lactational CDD also led only to a decrease TAS (-24%, p<0.001) in the suckling rat brains of both sexes Mg²⁺-ATPase activities showed no changes after any of the experimental procedures. Exp procedure 2: The administration of CDD led to a statistically significant decrease of the rat brain TAS (-29%, p<0.001 and to a significant increase of both AChE (+20%, p<0.001) and (Na⁺, K⁺)-ATPase (+35%, p<0.001) activities Mg²⁺-ATPase activity was found unaltered. Equilibrated diet, administered to early age CDD-treated rats of both sexes for an additional period of 18 weeks, restored the above parameters to control levels. Moreover, the in vitro experiments showed that Hcy could simulate these changes (at least under the examined in vitro conditions), while both Phe and Ala act protectively against the CDD-induced effects on the examined rat brain enzyme activities. The effects of early age CDD-feeding on the examined parameters are proved to be reversible through restoration to equilibrated diet, while our data suggest a role for Hcy (as a causative parameter for the CDD-induced effects) and a possible protective role for Phe and Ala (in reversing the observed CDD-induced effects). Exp Proc 3: There is a significant decrease between the two groups, CD and Control group, of the time spent on exploration of the new object, with a reduction of time for the group CD and a discrimination index D:-0,31±0,43. No significant deferences were observed between both sexes of rats in the main groups. Conclusions: Our data suggests that the reduced enzyme activities of the newborn CD brains were restored to normal after 21 days of either normal or CD lactation, possibly due to novel synaptogenesis, endogenous neuroregulation, and/or other substances acquired by lactation. Longer CDD-administration (during gestation, lactation and for three more weeks) leads to a statistically significant decrease in the offspring rat brain TAS and to a significant increase in both rat brain AChE and (Na+,K+)-ATPase activities restored into the control levels after equilibrated diet for 18 weeks. Moreover, the in vitro experiments showed that Hcy could simulate these changes (at least under the examined in vitro conditions), while both Phe and Ala act protectively against the Hcy-induced effects on the examined rat brain enzyme activities (indicating a possible role of these amino acids within the beneficial effects of equilibrated diet on the CDD-induced toxicity). The significant decrease for the group CDD of the time spent on exploration of the new object compared to the Control group in the ORT was found suggest that choline availability is important for the development of the brain during the first embryonic days.
