Η παρούσα διδακτορική διατριβή χωρίζεται σε τρεις κύριες θεματικές ενότητες.Στο πρώτο μέρος περιγράφεται η εκχύλιση του Hypericum triquetrifolium. Επίσης, περιγράφεται η απομόνωση της hyperibine J (γνωστή ως αδυπερφιρίνη), μιας ακυλο-φλωρογλυκινόλης τύπου Α, της οποίας η δομή που της είχε αποδοθεί ήταν λανθασμένη. Στο συγκεκριμένο μέρος αναλύεται ο τρόπος εύρεσης - αναθεώρησης της προτεινόμενης δομής της αδυπερφιρίνης σε hyperibine J.Στο δεύτερο μέρος περιγράφεται η εκχύλιση του Hypericum perforatum και η απομόνωση της υπερφορίνης και άλλων δευτερογενών μεταβολιτών του. Από τα επιμέρους κλάσματα της εκχύλισης διαπιστώθηκε η ύπαρξη ενός νέου μεταβολίτη, του μεθυλεστέρα του 3β-δεκαεπτανυλοξυ-ολεαν-12-εν-28-ικού οξέος. Επίσης, συντέθηκε η δεοξυκοχουμουλόνη και τα ακετυλιωμένα ανάλογα της. Τέλος, αξιολογήθηκε η αποτελεσματικότητά της υπερφορίνης, της δεοξυκοχουμουλόνης, των συντιθέμενων ακετυλιωμένων παράγωγων τους καθώς και των κλασμάτων της εκχύλισης, κατά των προνυμφών κουνουπιών του είδους Cx. pipiens. Στο τρίτο μέρος περιγράφεται ο σχεδιασμός, η σύνθεση και η βιολογική αξιολόγηση σε καρκινικές σειρές αναλόγων της υπερφορίνης. Τέλος, συνετέθη ένας νέος βιοδραστικός ανθρακικός σκελετός (δικυκλο[3.3.0]οκτανιο), η δομή του οποίου απεδείχθη με κρυσταλλογραφία ακτινών Χ.
The present thesis comprises of three major thematic units.In the first part, the structure elucidation of hyperibine J, originally assigned as adhyperfirin is described. Hyperibine J belongs to the polycyclic polyprenylated acylphloroglucinols (PPAPs) family and it is very similar to hyperforin, possessing a methyl substituent instead of a prenyl one on C-1. This phloroglucinol was isolated from the fresh aerial parts of Hypericum triquetrifolium and the revision of the molecule’s structure was based on spectroscopic analyses and chemical transformation to hyperibone J. In the second part, the extraction of Hypericum perforatum and isolation of hyperforin and other secondary metabolites are described. The existence of a new metabolite, methyl 3β-heptadecanoyloxy-olean-12-en-28-oate was detected for the first time. Also, the larvicidal effect of hyperforin, a bioactive compound of Hypericum perforatum, and deoxycohumulone (biosynthetic precursor of hyperforin) were evaluated against Culex pipiens (Diptera: Culicidae) for the first time. All the acetate analogues of hyperforin and deoxycohumulone were also synthesized and bioassayed to provide information on structural requirements for the tested compounds.In the third part, the design and the synthesis of hyperforin’s analogues were described. Finally, a novel synthesized bioactive carbon skeleton (bicyclo [3.3.0] octane system) was presented.
