      "@value" : "Η παρούσα διπλωματική εργασία εστιάζει στην παραγωγή βιοαιθανόλης από στέλεχος αραβοσίτου (corn stover) και στη βελτίωση των επιμέρους σταδίων της διεργασίας. Στο πρώτο μέρος της εργασίας μελετήθηκε η παραγωγή βιοαιθανόλης από στέλεχος αραβοσίτου με διαφορετικές προκατεργασίες και σε συνθήκες υψηλής συγκέντρωσης στερεών (high gravity). Εκπονήθηκαν πειράματα με τρία υλικά τα οποία είχαν προκατεργαστεί με τη μέθοδο έκρηξης ατμού- steam explosion (Δείγματα Α-Γ) και ένα υλικό που είχε προκατεργαστεί με τη μέθοδο organosolv (Δείγμα Δ). Τα δείγματα που είχαν υποστεί έκρηξη ατμού διέφεραν στις συνθήκες της προκατεργασίας και ειδικότερα στη συγκέντρωση προστιθέμενου H2SO4 (Δείγμα Α: 0% H2SO4, Δείγμα Β: 0.2% H2SO4, Δείγμα Γ: 1.0% H2SO4).  Στόχος των πειραμάτων ήταν αφενός η επιλογή της βέλτιστης εκ των συνθηκών κατά την έκρηξη ατμού (Δείγματα Α-Γ) και, αφετέρου, η σύγκριση της αποτελεσματικότητας των δύο μεθόδων προκατεργασίας (Δείγματα Α-Γ και Δ). \nΟ κύριος άξονας της εργασίας περιλαμβάνει ανάλυση σύστασης, ανάλυση κρυσταλλικότητας, δοκιμαστική ενζυμική υδρόλυση σε μικρή κλίμακα και ζύμωση σε μικρή κλίμακα, ώστε να διαπιστωθούν οι βέλτιστες συνθήκες κάθε σταδίου της διεργασίας, αλλά και να επιλεγούν τα υλικά με την καλύτερη συμπεριφορά. Στη συνέχεια, για τα δείγματα που επιλέχθηκαν από τις σειρές πειραμάτων ακολούθησε υδρόλυση και ζύμωση σε υψηλές συγκεντρώσεις στερεών (high gravity), με στόχο την επίτευξη υψηλής συγκέντρωσης αιθανόλης στο μίγμα της ζύμωσης. \nΑρχικά, η ανάλυση σύστασης των υλικών (πρωτόκολλο NREL) υπέδειξε απομάκρυνση λιγνίνης στο Δείγμα Δ και απομάκρυνση ημικυτταρίνης στα δείγματα που κατεργάστηκαν με έκρηξη ατμού,  με τη μεγαλύτερη απομάκρυνση να παρουσιάζεται στο Δείγμα Γ λόγω της υψηλής συγκέντρωσης προστιθέμενου οξέος. Παράλληλα, παρατηρήθηκε υποβάθμιση στο Δείγμα Γ πιθανότατα λόγω υδρόλυσης της κυτταρίνης κατά την προκατεργασία. Η ανάλυση κρυσταλλικότητας των υλικών πραγματοποιήθηκε με περίθλαση ακτίνων X και υπέδειξε αύξηση κρυσταλλικότητας των προκατεργασμένων υλικών σε σχέση με την πρώτη ύλη (CrIπρώτης ύλης= 55.5, CrIΑ= 59.8, CrIΒ= 57.7, CrIΓ= 64.8, CrIΔ= 62.1), με τη μεγαλύτερη αύξηση να εμφανίζεται στα υλικά από όπου απομακρύνθηκε μεγαλύτερο ποσοστό ημικυτταρίνης ή/και λιγνίνης. \nΑκολούθησε ενζυμική υδρόλυση μικρής κλίμακας σε συμβατικό σύστημα ανάδευσης (κωνικές φιάλες) με χαμηλή συγκέντρωση στερεών (10% w/v), όπου ελέγχθηκε η επίδραση της διαφορετικής συγκέντρωσης ενζύμου στην υδρόλυση της βιομάζας συναρτήσει του χρόνου (enzyme loading) για όλα τα υλικά. Παρατηρήθηκε υψηλότερη απελευθέρωση γλυκόζης στα Δείγματα Β και Δ, με τις μέγιστες συγκεντρώσεις γλυκόζης στο πέρας της υδρόλυσης (72h) να διαμορφώνονται ως εξής: Α (49.1 ± 0.99 g/L), Β (53.2 ± 1.23 g/L), Γ (41.7 ± 0.62 g/L), Δ (59.4 ± 0.45 g/L). Οι μέγιστες αποδόσεις υδρόλυσης βάσει της θεωρητικής απελευθερούμενης ποσότητας γλυκόζης κυμαίνονταν στο διάστημα 74%90%, μέσα στα πλαίσια των τιμών που αναφέρονται στη βιβλιογραφία. Επιλέχθηκε, τέλος, \n  \nη συγκέντρωση 12 mg/g DM ως βέλτιστο ενζυμικό φορτίο, η οποία συμβιβάζει την αποτελεσματικότητα της υδρόλυσης και το κόστος του προστιθέμενου ενζύμου. \nΣτη συνέχεια, έλαβε χώρα ζύμωση μικρής κλίμακας με τον μικροοργανισμό Saccharomyces cerevisiae σε κωνικές φιάλες και συγκέντρωση στερεών 10%w/v για τα Δείγματα Α-Δ. Τα υλικά ζυμώθηκαν στις βέλτιστες συνθήκες ενζυμικού φορτίου και χρόνου προϋδρόλυσης που είχαν διαπιστωθεί από τις προηγούμενες σειρές πειραμάτων. Οι μέγιστες συγκεντρώσεις αιθανόλης που επιτεύχθηκαν για τα δείγματα Α-Δ είναι: Α (21.2 g/L), Β (22.7 g/L), Γ (18.4 g/L) και Δ (15.4 g/L). Η χαμηλή συγκέντρωση παραγόμενης αιθανόλης στο Δείγμα Δ, παρά την καλή συμπεριφορά του κατά την υδρόλυση, αποδόθηκε στην πιθανή παρουσία κάποιου παρεμποδιστή της ζύμωσης στο υδρόλυμα. \nΑπό τα προηγούμενα πειράματα μικρής κλίμακας, επιλέχθηκε να μελετηθούν τα Δείγματα Β και Δ σε συνθήκες high gravity (24% w/w), σε μη συμβατικό σύστημα ανάδευσης (αναδευτήρας ελεύθερης πτώσης/ free fall mixer). Διερευνήθηκε αρχικά η απελευθέρωση γλυκόζης κατά την προϋδρόλυση, με τις τιμές συγκέντρωσης για τις 12h και τις 24h να είναι: Β (12h: 130.5 g/L, 24h: 148.3 g/L), Δ (12h: 118.6 g/L, 24h: 141.1 g/L). Κατά τη ζύμωση των υδρολυμάτων, μελετήθηκε η επίδραση της προσθήκης επιπλέον ποσότητας κυτταρινολυτικού ενζύμου για το Δείγμα Β (-/+). Οι μέγιστες συγκεντρώσεις αιθανόλης για το Δείγμα Β ήταν: 12h(-): 64.9 ± 2.96 g/L, 12h(+): 66.5 ± 1.06 g/L, 24h(-): 75.5 ± 4.93 g/L, 24(+): 71.5 ± 1.63 g/L. Το Δείγμα Δ στο ενζυμικό φορτίο 9 mg/g DM  παρουσίασε μικρότερη συγκέντρωση αιθανόλης, 64.5 ± 1.28 g/L. Οι συγκεντρώσεις αιθανόλης που επιτεύχθηκαν και στα δύο υλικά είναι από τις υψηλότερες που εμφανίζονται στη βιβλιογραφία για λιγνινοκυτταρινούχα βιομάζα.  \nΤέλος, πραγματοποιήθηκε υδρόλυση σε διαφορετικές συγκεντρώσεις αρχικών στερεών (6% w/v, 8% w/v, 10% w/v, 12% w/v) ώστε να διαπιστωθεί η επίδραση στην ενζυμική υδρόλυση. H απόδοση της υδρόλυσης στα συμβατικά συστήματα μειώθηκε σχεδόν γραμμικά με την αύξηση της συγκέντρωσης στερεών, ενώ αυξήθηκε ραγδαία στις υψηλές συγκεντρώσεις στερεών του συστήματος αναδευτήρα ελεύθερης πτώσης. Αποδείχθηκε έτσι η σημασία του συστήματος ανάδευσης για την επιτυχία της διεργασίας. \nΣτο δεύτερο μέρος της εργασίας, πολλαπλασιάστηκε και εκφράστηκε γονίδιο GH30 με πιθανή ξυλανολυτική δράση (putative xylanase). Στόχος της παραγωγής του καινοτόμου γονιδίου ήταν η διερεύνηση της συνεργιτικής δράσης που πιθανώς να έχει στην υδρόλυση της βιομάζας, ώστε να αυξηθεί ακόμα περισσότερο η απόδοση της συνολικής διεργασίας. Η έκφραση πραγματοποιήθηκε με συνθετικό, codon-optimized, γονίδιο του μικροοργανισμού Myceliophthora thermophila. Το γονίδιο πολλαπλασιάστηκε και υπέστη επεξεργασία με τεχνικές ανασυνδυασμένου DNA σε ξενιστή Escherichia coli TOP10.  Στη συνέχεια, προσαρτήθηκε σε πλασμίδιο pPICZαC, αυξήθηκε και εκφράστηκε σε ζύμη Pichia pastoris (στέλεχος Χ33). Η ενεργότητα του ενζύμου μελετήθηκε με τρυβλίο ελέγχου (Plate Assay) που περιείχε πιθανά υποστρώματα του."

      "@value" : "The present diploma thesis focuses on the production of bioethanol from corn stover and the improvement of the various stages of the production process. In the first part of the thesis\nthe production of bioethanol from corn stover was studied, pertaining to different pretreatment methods and in high gravity co\nnditions. Three steam -exploded samples (A-C)  and  an organosolv-pretreated  sample  were  tested.  Samples  A to C  differed  in  the conditions of pretreatment, regarding the added concentration of H2SO4 (Sample A: 0% H2SO4, Sample B: 0.2% \nH2SO4, Sample C: 1.0% H2SO4). It was, firstly, intended to choose the  best steam  explosion  conditions for  Samples  A-\nC  and, secondly, to  compare  the efficacy of both pretreatment methods (Samples A -C and D).The  main experimental focus  of \nthis work  includes biomass composition  analysis, crystallinity analysis, enzymatic hydrolysis on a small scale, and fermentation \non a small scale, in order to establish the optimal conditions of each process stage, and to select the samples with  the best \nbehavior. Afterwards,  the  samples  that  were selected  from  the \nabove series of experiments were hydrolyzed and fermented \nat high solids concentrations (high  gravity), in  order to  achieve  a  high  concentration  of  ethanol  in  the  fermentation broth.\nTo begin with, biomass composition analysis (NREL) indicated removal of lignin in Sample D and removal of hemicellulose in the samples treated by steam explosion.The greatest removal occurred in   Sample   C   because   of   its   high   concentration   of   added   acid. Furthermore, deterioration in the Sample C was observed probably due to hydrolysis of cellulose during the retreatment. Crystallinity analysis of materials was performed by X\n-ray  diffraction  and showed increased  crystallinity  of  the samples comparing to  the raw material (CrIraw= 55.5, CrIA\n= 59.8, CrIB= 57.7, CrIc= 64.8, CrID= 62.1). The largest increase \nappeared in the materials where a greater percentage of hemicellulose and/or lignin was removed. Furthermore,  enzymatic  hydrolysis on  a small  scale  in  a  conventional \nstirring system (flasks) with a low solids concentration (10% w/v)\nwas performed. The experiment tested the  effect  of  different  enzyme  concentrations in  the  hydrolysis  of  the  biomass  as  a \nfunction of time (enzyme loading). There was a higher release of glucose in samples B and D, the maximum glucose oncentrations (at 72h) being as follows: A (49.1 ± 0.99 g/L), B (53.2 ± 1.23 g/L), C (41.7 ± 0.62 g/L), D (59.4 ± 0.45 g/L). The maximum hydrolysis yields -based  on  the  theoretical  amount  of  glucose  released-\nranged  in  the  interval  74%-90% (within  the  values reported  in  the  literature). Finally, the  concentration  of 12 mg/g  DM was chosen as the optimal enzyme load, which compromises the efficiency of hydrolysis and the cost of the added enzyme. Moreover, fermentation was held on  a small  scale  in  flasks \nusing the  microorganism Saccharomyces  cerevisiae and \nin 10%  w/v solids  concentration. The  materials  were treated\nin the optimal enzyme load that had been established by the previous series of experiments.  The  maximum  ethanol  concentrations  obtained  for  samples  A-D were:  A (21.2 g/L),  B  (22.7 g/L),  C  (18.4 g/L)  and  D  (15.4 g/L).  The  low  concentration  of  ethanol produced in Sample D, despite its \ngood behavior upon hydrolysis, was attributed to the possible presence of a fermentation inhibitor in the hydrolysate. From the \nabove small-scale experiments, samples B and D were selected to be studied in high gravity conditions (24% w/w), in a non-conventional stirring system (free fall mixer). The glucose\nconcentration values for 12h and 24h pre-hydrolysis we re: B (12h: 130.5 g/L, 24h:  148.3  g/L),  D  (12h:  118.6  g/L,  24h:  141.1  g/L).  During  the  fermentation  of the hydrolyzate, the effect of adding an additional quantity of cellulolytic enzyme \nwas studied for Sample B (-/+). The maximum concentrations of ethanol for Sample B were: 12h (-): 64.9 ± 2.96 g/L, 12h (+): 66.5 ± 1.06 g/L, 24h (-): 75.5 ± 4.93 g/L, 24 (+): 71.5 ± 1.63 g/L. \nSample  D  showed a  lower  concentration  of  ethanol: 64.5 ± 1.28 g/L. The  ethanol concentrations achieved in both materials are among the highest displayed in literature of lignocellulosic biomass. Finally, hydrolysis at different solids concentrations (6% \nw/v, 8% w/v , 10% w/v, 12% w/v) was tested to determine the effect on enzyme hydrolysis. The hydrolysis yield decreased almost  linearly with the increasing  concentration  of  solids  in conventional  systems, but grew  rapidly  at  high  solids  concentrations  of  the  free  fall  mixer  system. Thus  the \nimportance of the stirring system on the efficacy of the process \nwas demonstrated. In the second part of the present thesis, \na GH30(CAZy) gene with a putative xylanolytic action was \nmultiplied  and  expressed.  The aim of  the novel  enzyme\nproduction was  to investigate the synergistic effect that it may\nhave on the hydrolysis of biomass, to increase further\nthe  efficiency  of  the  overall  process.  The  expression  was  carried  out  with  a synthetic, codon-optimized gene of the microorganism Myceliophthora thermophila. The gene  was  amplified  and  treated with DNA  techniques,  and  was  inserted  in the host microorganis m Escherichia coli (strain TOP10. It was then attached to the plasmid pPICZaC and expressed in Pichia pastoris cells (strain X33). The activity of the enzyme was studied \nby Plate Assay, with a plate containing its potential substrates."
