Given that the δ-opioid receptor (δ-OR) contains the same \nstructural motif within its C-terminal tail (δ-CT), we have shown that STAT5Β \ninteracts also with this tetra-peptide of the δ-CT. Agonist exposure of HEK293 \ncells, stably expressing the flag-δ-OR, led to a G protein-dependent STAT5Β \nphosphorylation mediated by c-Src kinase. Additional studies indicated that δ-\nOR serves as a platform for the formation of a multi-component signaling \ncomplex, consisting of STAT5B, c-Src kinase, Gβγ and selective Gαi/o protein \nsubunits. Furthermore, our findings demonstrated that δ-OR activation leads to \nneuronal cell survival and neurite outgrowth via the aforementioned signaling \npathway involving Gαi/o proteins and phosphorylated STAT5B. Προηγούμενες μελέτες του εργαστηρίου μας έδειξαν ότι το \nσυντηρημένο YXXL μοτίβο στο καρβοξυτελικό άκρο του μ-οπιοειδούς υποδοχέα (μ-OR) \nαποτελεί σημείο αγκυροβόλησης για τον μεταγραφικό παράγοντα STAT5A, ο οποίος \nφωσφορυλιώνεται μετά την ενεργοποίηση του μ-OR. Δεδομένου ότι και ο δ-οπιοειδής \nυποδοχέας (δ-OR) περιέχει το ίδιο δομικό μοτίβο στο καρβοξυτελικό του άκρο (δ-\nCT), αποδείξαμε ότι η STAT5Β αγκυροβολεί σε αυτή την περιοχή του δ-CT. Έκθεση \nτων HEK293 κυττάρων, που εκφράζουν τον flag-δ-OR, σε δ-οπιοειδείς αγωνιστές \nοδηγεί στη Gαi/o-εξαρτώμενη φωσφορυλίωση της STAT5Β μέσω μιας c-Src τυροσινικής \nκινάσης.
