      "@value" : "Σκοπός της παρούσας μελέτης ήταν διερεύνηση της ικανότητας πολυλειτουργικών \nνανοφορέων πολυ(D,L-γαλακτικού–co-γλυκολικού οξέος) (poly \n(D,L-lactic-co-glycolic acid, PLGA) να επάγουν προστατευτικές αποκρίσεις σε \nπρότυπο σπλαγχνικής λεϊσμανίασης. Σε νανοφορείς PLGA ενσωματώθηκε εκχύλισμα των \nπρομαστιγωτών μορφών του παρασίτου L. infantum (sLiAg) και το ανοσοενισχυτικό \nMPLA. Οι νανοφορείς PLGA τροποποιήθηκαν επιφανειακά με ένα 8-πεπτίδιο (p8), η \nαλληλουχία του οποίου παρουσιάζει ομολογία με την περιοχή πρόσδεσης του TNFα \nστους υποδοχείς του. \nΟι νανοφορείς με την επιφανειακή τροποποίηση προσλαμβάνονταν πιο αποτελεσματικά \nαπό τα δενδριτικά κύτταρα, γεγονός που αποδίδεται σε ενδεχόμενη στόχευση των \nδενδριτικών κυττάρων από το p8, αλλά και στη μείωση της απόλυτης τιμής του \nεπιφανειακού φορτίου των νανοφορέων. Η ενσωμάτωση του sLiAg ή και του MPLA, \nκαθώς και η επιφανειακή τροποποίηση με το p8 προκάλεσαν την ωρίμανση και τη \nλειτουργική διαφοροποίηση των δενδριτικών κυττάρων, τα οποία είχαν την \nικανότητα να παρουσιάζουν τα αντιγόνα του παρασίτου σε Τ λεμφοκύτταρα επάγοντας \nτην ειδική ενεργοποίησή τους. Ο εμβολιασμός ποντικών BALB/c με τους νανοφορείς \nPLGA-sLiAg-MPLA και p8-PLGA-sLiAg είχε ως αποτέλεσμα την ανάπτυξη διαμένουσας \nπροστασίας έναντι της μόλυνσης με L. infantum, μέσω της ενεργοποίησης \nβοηθητικών και κυτταροτοξικών Τ λεμφοκυττάρων και της καταστολής των \nυποπληθυσμών ΤΗ2 και Τ ρυθμιστικών λεμφοκυττάρων, προτείνοντας τους \nνανοσχηματισμούς αυτούς ως υποψήφια εμβόλια έναντι της σπλαγχνικής \nλεϊσμανίασης."

      "@value" : "The aim of the present study was to investigate the ability of \npoly(lactic-co-glycolic) acid (PLGA) nanoparticles (NPs) to induce protection \nagainst experimental visceral leishmaniasis. Nanoformulations based on PLGA NPs \nwere synthesized and encapsulated a crude mix of proteins from Leishmania \ninfantum promastigotes (sLiAg) and MPLA, as adjuvant. PLGA NPs were \nsurface-modified with a synthetic 8-peptide mimicking the TNFα docking region, \nin order to enhance dendritic cell targeting. \nSurface modified PLGA NPs were taken up more efficiently from dendritic cells \ncompared to non-modified NPs. This is attributed to possible enhancement of \ndendritic cell targeting, as well as to the reduction of the absolute value of \nz potential. Incorporation of sLiAg and/or MPLA and surface modification with \np8 induced the maturation and functional differentiation of dendritic cells \nresulting in their enhanced ability of presenting parasitic antigens and \nactivating in vitro T cells. Vaccination of BALB/c mice with PLGA-sLiAg-MPLA or \np8-PLGA-sLiAg NPs induced long-lasting protection against L. infantum infection \ndue to activation of T helper and cytotoxic T cell populations and simultaneous \nsuppression of TH2 and regulatory T cell subpopulations. Based on the above \nresults, the nanoformulations used are suggested as candidate vaccines against \nvisceral leishmaniasis."
