      "@value" : "Στόχος της παρούσας εργασίας ήταν η διερεύνηση του ρόλου της εκκρινόμενης α-\nσυνουκλεΐνης (ΑΣΥΝ) στην ομοιόσταση των νευρικών κυττάρων. Η ΑΣΥΝ συνδέεται \nγενετικά και βιοχημικά με την παθογένεια της νόσου του Parkinson. Προκειμένου \nνα εξετάσουμε πιθανούς μηχανισμούς μέσω των οποίων η ΑΣΥΝ συμβάλλει στο \nνευροεκφυλισμό, δημιουργήσαμε μία ανθρώπινη κυτταρική σειρά, στην οποία η \nφυσικού τύπου ανθρώπινη ΑΣΥΝ εκφράζεται σε υψηλά επίπεδα. Δείξαμε ότι η ΑΣΥΝ \nεκκρίνεται φυσιολογικά στο θρεπτικό μέσο των κυττάρων, μέσω ενός μη κλασικού \nμηχανισμού, ο οποίος περιλαμβάνει, εν μέρει, τη σύνδεσή της με εξωσώματα. Οι \nεκκρινόμενες μορφές της ΑΣΥΝ προκαλούν νευροεκφυλισμό, χωρίς, ωστόσο, να \nενδοκυτταρώνονται. Σημειωτέον, η εκκρινόμενη ΑΣΥΝ αυξάνει τη ρευστότητα της \nπλασματικής μεμβράνης των κυττάρων-δεκτών και την εισροή ιόντων ασβεστίου,  \nμέσω των L- και N- τύπου τασεοελεγχόμενων διαύλων ασβεστίου. Επιπλέον, το \nπλεονάζον ασβέστιο συσσωρεύεται στα μιτοχόνδρια. Να σημειωθεί ότι οι παραπάνω  \nδιαταραχές  συμβαίνουν πριν τα σημάδια του κυτταρικού θανάτου γίνουν εμφανή. Σε \nμεταγενέστερο χρονικό διάστημα, το οποίο συμπίπτει με την κορύφωση του \nεκφυλισμού, διαπιστώθηκε σημαντική αύξηση της ενεργοποίησης των καλπαϊνών, \nπρωτεασών που πυροδοτούν καταρροϊκά μονοπάτια θανάτου. Τέλος, η χορήγηση \nειδικών φαρμακολογικών παραγόντων, που είτε παρεμποδίζουν την είσοδο του \nασβεστίου στα κύτταρα είτε δεσμεύουν το εξωκυττάριο ή ενδοκυττάριο ασβέστιο, \nμειώνει τη νευροτοξική επίδραση της εκκρινόμενης ΑΣΥΝ."

      "@value" : "In the current study we addressed the role of secreted a-synuclein (AS) on \nneuronal homeostasis. AS has been genetically and biochemically linked to \nParkinson’s disease pathogenesis. In order to decipher possible mechanisms via \nwhich extracellular AS elicits its pathogenic functions, we created a human \ncell-line, in which human wild type AS is expressed in high levels. We found \nthat AS can be physiologically secreted in the culture medium of the \noverexpressing cells, via an unconventional secretory pathway, which partly \nrelies on exosomes. Moreover, secreted forms of AS lead to neurodegeneration, \nvia a pathway that does not include internalization of the protein.  \nImportantly, the presence of secreted AS results in increased plasma membrane \nfluidity, augmented calcium entry via the L- and N- type voltage-operated \ncalcium channels and preferential sequestration of the excessive calcium into \nthe mitochondria. Interestingly, the described events precede AS-induced \nneurotoxicity. At a later time point, which corresponds to the peak of \nAS-evoked neuronal death, we observed significant activation of calpains, \ncalcium-dependent proteases which trigger downstream death cascades. Lastly, by \nthe use of specific chemical compounds, which either block calcium entry or \nchelate intracellular or extracellular calcium, we were able to spare cells \nfrom secreted AS adverse effects."
