      "@value" : "Ιn the adult mammalian brain neural\nstem/progenitor cells (NPCs) have a spatially restricted distribution, and\nneurogenesis takes place only in specific brain areas. Thus, all brain regions \nbeyond\nthe SVZ and dentate gyrus are considered “non-neurogenic”. Recently, it is \nproposed\nthat a low level of neurogenesis occurs also in “non-neurogenic regions”, such \nas the\nstriatum and, to a lesser extent, the cortex. Importantly, brain lesions have\nsucceeded to unmask neurogenic activity in such regions in laboratory rodents. \nIn\nthis study we aim to investigate whether disturbance of the adult mouse SVZ,\nactivates cells of neighboring non-neurogenic regions to acquire a neural \nprogenitor\npotential. To this end, we have performed a protocol aiming to reduce the \nSVZ-stem\ncell population, using a combination of mechanical trauma in the SVZ area, to\nactivate quiescent or slowly proliferating NPCs, followed by subsequent \nrepeated,\nstereotaxic intraventricular injections of the mito-toxic agent arabinoside-C \n(Ara-C),\nthat kills rapidly proliferating NPCs. \nFinally we sacrifice the animals s 6 weeks after the primary trauma.\nThe results of this study support the hypothesis that the injection of\nmitotoxic AraC in the lateral ventricles is responsible for the the disruption \nof the\nSVZ and the induction of the ectopic presence of newborn neurons in \nnonneurogenic\nareas.The\nfact that several mitotoxic factors are widely used as chemotherapeutic agents \nand\nthat patients undergoing chemotherapy exhibit progressive cognitive deficits, \nraises\nthe question whether these treatments lead to a disruption of neurogenesis.\nTherefore, further study of this SVZ-disorganization model will shed light into\nimportant questions related to NPCs and neighboring parenchymal brain cells’\nresponse to chemotherapeutic treatment."

      "@value" : "Τα ενήλικα νευρικά βλαστικά (ΝΒΚ) και πρόδρομα νευρικά\nκύτταρα έχουν ορισμένη χωροταξική κατανομή και η νευρογένεση λαμβάνει χώρα σε \nαυστηρά καθορισμένες περιοχές του εγκεφάλου. Υπό αυτό το πρίσμα όλες οι\nυπόλοιπες περιοχές εκτός από την υποκοιλιακή ζώνη (SVZ) και την οδοντωτή έλικα \nτου ιππόκαμπου θεωρούνται «μη νευρογενετικές». Εντούτοις, πρόσφατα έχει \nπαρατηρηθεί ότι η εγκεφαλική βλάβη σε μη νευρογενετικές περιοχές όπως ο φλοιός \nκαι το ραβδωτό σώμα ενεργοποιεί τη νευρογενετική δραστηριότητα διαφοροποιημένων \nκυττάρων στην περιοχή της βλάβης. Στη μελέτη αυτή στόχος είναι να διερευνηθεί \nκατά πόσον η διατάραξη της φωλεάς και η μείωση των ΝΒΚ στη SVZ ενήλικου \nποντικού ενεργοποιεί κύτταρα των γειτονικών μη νευρογενετικών περιοχών ώστε να \nαποκτήσουν δυναμικό πρόδρομων νευρικών κυττάρων. Για το σκοπό αυτό, έχουμε \nπραγματοποιήσει ένα πρωτόκολλο με στόχο τη μείωση του πληθυσμού των βλαστικών \nκυττάρων της SVZ, χρησιμοποιώντας έναν συνδυασμό μηχανικού τραύματος της SVZ, \nώστε να ενεργοποιηθούν τα αργά πολλαπλασιαζόμενα ΝΒΚ, ακολουθούμενο από \nεπαναλαμβανόμενες στερεοταξικές\nεγχύσεις του μιτοτοξικού παράγοντα κυτοσίνη αραβινοσίδη (Ara-C), που σκοτώνει \nτα ταχέως πολλαπλασιαζόμενα ΝΒΚ, στις κοιλίες του εγκεφάλου. Τα ως τώρα \nδεδομένα της μελέτης αυτής, υποστηρίζουν πως η έγχυση του μιτοτοξικού παράγοντα \nείναι υπεύθυνη για τη διατάραξη της SVZ και την επαγωγή της εκτοπικής παρουσίας \nνέων νευρώνων σε μη νευρογενετικές περιοχές. Το γεγονός πως διάφοροι \nμιτοτοξικοί παράγοντες χρησιμοποιούνται ευρέως ως χημειοθεραπευτικές ουσίες και \nπως σε ασθενείς που\nυπόκεινται σε χημειοθεραπείες παρατηρούνται προοδευτικά γνωσιακά ελλείμματα, \nεγείρει το ερώτημα κατά πόσο αυτές οι θεραπείες οδηγούν σε διατάραξη της \nνευρογένεσης. Για το λόγο αυτό περαιτέρω μελέτη του συγκεκριμένου μοντέλου\nδιατάραξης της SVZ μπορεί να δώσει απάντηση σε σημαντικά ερωτήματα που αφορούν \nστην απόκριση των ΝΒΚ, καθώς και διαφοροποιημένων κυττάρων γειτονικών περιοχών \nτου εγκεφάλου μετά από χημικοθεραπευτική αγωγή."
