Οι \nπαθογενετικοί μηχανισμοί δεν έχουν πλήρως αποσαφηνιστεί, όμως τα εμπλεκόμενα \nαυτο-αντισώματα έχουν προταθεί να ενεργοποιούν διάφορους κυτταρικούς πληθυσμούς \n(αιμοπετάλια, ενδοθηλιακά κύτταρα, λευκοκύτταρα) προσδίδοντάς τους έναν \nθρομβοφιλικό φαινότυπο. Στην παρούσα μελέτη μετρήθηκαν μέσω ανοσοενζυμικής \nμεθόδου (ELISA) κυκλοφορούσες ουσίες [αιμοπεταλιακός παράγων 4 (PF4), \nP-σελεκτίνη, μεταλλοπρωτεϊνάσες της εξωκυττάριας θεμέλιας ουσίας (MMPs) -1, -3, \n-9] που αποτελούν δείκτες ενεργοποίησης των ενεχόμενων στο ΣΑΦ κυτταρικών \nπληθυσμών. Μεταξύ των ασθενών με ΣΑΦ, αυτοί με υποτροπιάζοντα θρομβωτικά \nεπεισόδια (n=16) παρουσιάζουν σημαντικά υψηλότερες τιμές PF4 πλάσματος από \nαυτούς με μεμονωμένα επεισόδια (n=18) και τους υγιείς δότες (Υ.Δ) (n=22). \nΕπίσης, τα επίπεδα P-σελεκτίνης πλάσματος βρέθηκαν σημαντικά υψηλότερα στους \nασθενείς με πρωτοπαθές ΣΑΦ (n=14) έναντι των Υ.Δ (n=24), των ασθενών με \nδευτεροπαθές ΣΑΦ (δΣΑΦ) παρουσία Συστηματικού Ερυθηματώδους Λύκου (ΣΕΛ) (n=22) \nκαι των ασθενών με ΣΕΛ (n=9). Η λήψη ασπιρίνης μειώνει σημαντικά τα επίπεδα \nP-σελεκτίνης στο δΣΑΦ. Τα ευρήματά μας \nτονίζουν τη σημασία της ενεργοποίησης των αιμοπεταλίων και του ενδοθηλίου στην \nπαθογένεση του ΣΑΦ." }, { "@language" : "en", "@value" : "Antiphospholipid Syndrome (APS) is an autoimmune thrombophilia characterized by \nthromboembolism and/or pregnancy morbidity. The underlying pathogenetic \nmechanisms are yet unclear, but evidence exists that the circulating \nautoantibodies (antiphospholipid antibodies, aPL) activate different cell \npopulations (platelets, endothelial cells, leukocytes) inducing a \nprocoagulant/proinflammatory phenotype. This study examined the levels of \ndifferent blood-borne soluble markers [Platelet Factor 4 (PF4), P-selectin, \nmatrix metalloproteinases (MMPs) -1, 3-, -9] of APS involved cellular \nactivation using enzyme-linked immunosorbent assays (ELISA). Plasma PF4 levels \nwere significantly higher in our group of APS patients with recurrent \nthrombosis (n=16) than those with a sole event (n=18) and healthy donors (HD) \n(n=22). Plasma P-selectin levels were significantly increased in primary APS \npatients (n=14) compared with HD (n=24), Systemic Lupus Erythematosus (SLE)/APS \npatients (n=22) and SLE patients (n=9). SLE/APS patients treated with aspirin \ndisplayed significantly lower P-selectin levels. No difference in MMP-1 and \nMMP-9 levels was found between APS patients and HD, while the difference \nobserved in MMP-3 levels is probably due to the influence of corticosteroids. \nFinally, there was no correlation between the levels of the soluble markers \nstudied and the titer of aPL.
