      "@value" : "Ο καρκίνος της ουροδόχου κύστεως αποτελεί το δεύτερο σε συχνότητα και πρώτο σε \nθνησιμότητα καρκίνο του ουρογεννητικού συστήματος, με την επιθετικότητά του να \nσχετίζεται με έκφραση της πρωτεΐνης προφιλίνης-1. Η προφιλίνη-1 δεσμεύει τα \nμονομερή της β-ακτίνης και ρυθμίζει τον πολυμερισμό της ως απόκριση σε \nεξωκυτταρικά σήματα. Σκοπός της παρούσας ερευνητικής εργασίας είναι η μελέτη \nτης επίδρασης της πρωτεΐνης προφιλίνης-1 στη μεταστατική κυτταρική σειρά \nκαρκίνου ουροδόχου κύστεως, Τ24Μ, μέσω προσδιορισμού του δυναμικού εμφάνισης \nόγκων και μεταστατικότητας των καρκινικών κυττάρων με τη χρήση του γονιδίου \nαναφοράς της λουσιφεράσης. Αρχικά, δημιουργήθηκαν γενετικώς τροποποιημένα \nκύτταρα Τ24Μ με λεντιϊκούς φορείς, τα κατεσταλμένης προφιλίνης-1 Τ24Μ shPFN1 \nκαι τα κύτταρα ελέγχου Τ24Μ shSCRAMBLED. Μείωση στην έκφραση της προφιλίνης-1 \nστα κύτταρα T24M οδήγησε σε μειωμένο πολυμερισμό της ακτίνης, αναστολή της \nκυτταρικής κινητικότητας και της ικανότητας προσκόλλησης στην εξωκυτταρική \nουσία, αυξημένη παραγωγή γνωστών ογκο-κατασταλτικών παραγόντων και μείωση των \nεπιπέδων αγγειογενετικών πρωτεϊνών. Επιπλέον, η έλλειψη της προφιλίνης-1 \nοδήγησε σε μειωμένη αύξηση και μετάσταση των καρκινικών όγκων in vivo, μετά από \nεμβολιασμό των σημασμένων με λουσιφεράση καρκινικών κυττάρων σε NOD/SCID \nποντίκια. Συμπερασματικά προτείνεται ότι η προφιλίνη-1 έχει ενεργό ρόλο σε \nδιάφορες λειτουργίες των καρκινικών κυττάρων ουροδόχου κύστεως και κυρίως στη \nμεταναστευτική τους ικανότητα."

      "@value" : "Bladder cancer is the second in incidence and first in mortality cancer of the \ngenitourinary system and its aggressiveness has been correlated with the \nexpression of profilin-1. Profilin-1 binds monomeric-actin and regulates \npolymerization in response to extracellular signals. In this thesis, we \ntransduced the Τ24Μ cell line with a lentiviral vector that includes shRNA \nsequence complementary to profilin-1 (shPFN1) or shRNA non targeting sequence \n(shSCRAMBLED) οr a vector with the reporter gene of luciferase. Decrease of \nprofilin-1 expression in T24M cells resulted in decreased polymerization of \nactin and inhibition of cell motility and adhesion to extracellular matrix. \nFurthermore, decreased levels of profilin-1 were correlated with \ndown-regulation of factors related to angiogenesis, migration and metastasis \nand up-regulation of proteins with onco-suppressor activity. In addition, \ndecreased levels of cytoplasmic profilin-1 resulted in reduction οf tumour \nformation and metastasis in vivo, after injection of T24M cells that expressed \nthe reporter gene of firefly luciferase, in NOD/SCID mice. In conclusion, it is \nindicated that profilin-1 has an active role in important functions of bladder \ncancer cells. This protein could be an ideal prognostic marker of the \noccurrence of bladder cancer or a cancer marker for the transition from \nnon-invasive to invasive type of cancer."
