EAE represents an animal model for multiple \nsclerosis. In addition, in vitro studies have linked tolerance to lower \nantigen-specific proliferation and the development of peptide-specific T cell \nanergy, despite the fact that the expression of Th1 and Th17 effector cytokines \nwas not impaired1. The objective of this Thesis was to examine in vivo whether \nthe protective effects of mannan-cojugated peptides are mediated by changes in \nproliferation or trafficking of activated T cells into the CNS during EAE. \nChimeric mice were generated, in which MOG35-55(MOG)-specific T cells isolated \nfrom 2D2 MOG-specific T cell receptor transgenic mice2 that had previously been \nvaccinated with mannan-MOG or PBS control, were labeled with fluorescent CFSE \nand adoptively transferred into congenic (CD45.1) recipients with ongoing \nMOG-EAE.
