Πρόσφατες μελέτες \nέδειξαν ότι οι PC-1 και PC-2 συμμετέχουν σε σημαντικές κυτταρικές λειτουργίες, \nενεργοποιώντας ενδοκυττάρια μονοπάτια σηματοδότησης, μεταξύ των οποίων το \nμονοπάτι του JAK/STAT.\nΣκοπός της παρούσας μελέτης είναι η διερεύνηση της βιολογικής σημασίας της \nμηχανοευαίσθητης πρωτεϊνης, PC-1 σε ανθρώπινους οστεοβλάστες, συσχετίζοντας τον \nπιθανό ρόλο της στη μηχανική φόρτιση με τη ρύθμιση του σηματοδοτικού μονοπατιού \nJAK/STAT. \nΓια το σκοπό αυτό αρχικά, επιβεβαιώσαμε την έκφραση των PC-1 και PC-2 σε \nανθρώπινους οστεοβλάστες, με τεχνικές ανοσοφθορισμού και ανοσοστύπωσης Western. \nΣτη συνέχεια, επιβεβαιώσαμε την αλληλεπίδραση των πρωτεϊνικών μορίων PC-2, \nPC-1, και PC-1, pJAK2 μέσω της τεχνικής ανοσοφθορισμού Duolink. Ακολούθως, \nαποδείξαμε μέσω κυτταρομετρίας ροής την PC-1-μεσολαβούμενη ενεργοποίηση του \nσηματοδοτικού μονοπατιού JAK/STAT μετά από μηχανική διέγερση. Με τη μέθοδο \nTransAM, παρατηρήθηκε ενεργοποίηση του μεταγραφικού παράγοντα pSTAT3 μετά από \nμηχανική τάση.
