Thymbra\n2) Οίνο στον οποίο εκχυλίστηκε η ίδια δρόγη\nΑμφότερα προέκυψαν κάτω από ελεγχόμενες συνθήκες προκειμένου να προκύψουν προϊόντα με σταθερά και συγκεκριμένα χαρακτηριστικά. \nΧρησιμοποιήσαμε αδιαχώριστα τα υπέργεια μέρη (φύλλα, καρπούς, κλαδιά) της Satureja thymbra. Παρασκευάσαμε και αναλύσαμε τα εξής δείγματα: \nΑ) οινική εκχύλιση του Satureja thymbra σε συγκεντρώσεις 0 (μάρτυρας), 3, 6 & 9%\nΒ) προσθήκη συγκεκριμένης μάζας φυτού σε μούστο και συν-ζύμωση σε συγκεντρώσεις 0 (μάρτυρας), 3, 6 & 9% \nΓ) Διαλύματα πρότυπου μούστου και πρότυπου οίνου. \nΤα οργανικά φορτία των μεταβολιτών όλων των παραπάνω υδατοαλκοολικών δ/των (οινικών εκχυλίσεων, συνζυμώσεων, προτύπων και «τυφλών») δεσμεύτηκαν μετά από έκλουση από μακρόπορες, πολυμερικές μη-ιοντικές ρητίνες τύπου XAD. Όπως και στην πτυχιακή, χρησιμοποιήθηκαν 2 είδη ρητινών με διαφορετική χημεία και τεχνολογία πολυμερισμού (XAD-4 και XAD-7) και επομένως με διαφορές στις κατηγορίες μορίων που μπορούν να δεσμεύουν (κυρίως όσον αφορά στην πολικότητα των μορίων).\nΑπό προηγούμενη μελέτη έχουμε εντοπίσει το μέγιστο όριο «ανοχής» των ρητινών σε αλκοόλη, ώστε να μην αποδεσμεύονται οι μεταβολίτες κατά την διάρκεια των εκλούσεων (όλα τα δείγματα είναι υδατοαλκοολικά δ/τα, και η EtOH είναι ένας από τους δ/τες αποδέσμευσης (και αναγέννησης των πολυμερικών ρητινών). Έτσι, έχουμε καθορίσει την καλύτερη συμπεριφορά της XAD4 σε αραίωση του δείγματος στο 25% (25% οίνος και 75% H2O) ενώ στην XAD7 στο αναραίωτο προϊόν.\nΗ διαδικασία εν συντομία: \nα) διέλευση του δείγματος από τη ρητίνη (σε γυάλινες χρωματογραφικές στήλες) και συλλογή του διηθήματος το οποίο ονομάζουμε \"απόβλητο\"\nβ) έκπλυση με συγκεκριμένη ποσότητα H2O και συλλογή των εκπλύσεων\nγ) Αποδέσμευση από τη ρητίνη με συγκεκριμένη ποσότητα MeOH.\nδ) Ξήρανση των δειγμάτων.\nΤα δεσμευμένα φορτία όλων των εκχυλισμάτων εξετάστηκαν ως προς τις μάζες τους. Μετά την απαραίτητη διαδικασία του dereplication που διενεργήσαμε στα φάσματα LC-HRMS, διαπιστώθηκαν σημαντικές διαφορές μεταξύ των δεσμευμένων φορτίων των οινικών εκχυλισμάτων και των αντίστοιχων των συνζυμώσεων της S. Thymbra και για την XAD-4 και για την XAD-7." }, { "@language" : "en", "@value" : "The present thesis represents a study of the medical wine that originates from the fermentation of the Malagouzia grape variety (Vitis vinifera L. cv. Malagouzia) in the presence of the medical plant Satureja thymbra. \nWith the aid of the Agricultural University of Athens and the method of preparation of Medical Wines from Materia Medica we brought three products in our laboratory: 1) wine from co-fermentation of must and Satureja thymbra, 2) wine in which the same plant was extracted and 3) prototype must and wine solutions with the same quantity of plant material. Both wines occurred under totally controlled conditions in order to ensure similarity and stability in quality characteristics. We used above-ground parts of the plant Satureja thymbra for the preparation of the three series of products:\nA)\tco-fermentation of must and Satureja thymbra in 0%, 3%, 6% and 9% w/v\nB)\textraction of Satureja thymbra in the same w/v ratios\nC)\tprototype wine and must solutions, 9% w/v (called BLIND)\nThe organic metabolites of the above water/alcohol solutions (co-fermentation, wine extraction and BLIND) were held after their passing through two different (in chemical characteristics and polymerization technology) XAD type macropore polymeric resins (XAD4 & XAD7) that can, according to their specifications, bind different kinds of molecules (mostly according to polarity and molecular size of metabolites).\nFrom a previous work of ours we had set the limits of “resistance” of the resins in the presence of alcohol, so that the metabolites would (and could) be held in the resins despite the ~14% in EtOH concentration of wine since EtOH (and also MeOH) is an elution (and regenerative) solvent for the XAD type resins. These limits were set to 25% for XAD4 (25% wine-75% H2O) and 100% wine for XAD7. \nThe followed procedure in summary: \na)\tpassing of wine sample through the resin (in glass chromatographic columns) and collection of the filtrated solution (we called it WASTE)\nb)\trinsing with a certain quantity of H2O and collection of the filtration (we called it RINSING or RINSE)\nc)\telution of binded metabolites using a certain quantity of MeOH\nd)\tdrying of all collections\nAll dried samples were weighted, compared as of their weight and sent for LC-HRMS research.
