Δομικά, οι πρωτεΐνες Pkhd1l1 του zebrafish μοιάζουν με την ανθρώπινη πρωτεΐνη PKHD1 που προκαλεί αυτοσωμική υποτελή πολυκυστική ασθένεια των νεφρών. Υποθέσαμε πως τα γονίδια pkhd1l1 του zebrafish εμπλέκονται στην δημιουργία βλεφαρίδων. Για να εξετάσουμε την υπόθεση μας, δημιουργήσαμε διπλα pkhd1l1 zebrafish knockouts. Εξετάσαμε επίσης Pkhd1l1 knockout ποντίκια για πιθανές διαταραχές των κινούμενων βλεφαρίδων. Τα pkhd1l1 zebrafish knockouts έδειξαν μη φυσιολογικούς ωτόλιθους 24 ώρες μετά την γονιμοποίηση, αλλά κανένα άλλο σύμπτωμα που να σχετίζεται με προβλήματα κινούμενων βλεφαρίδων. Τα double pkhd1l1 zebrafish knockouts εμφάνισαν υπερέκφραση του βιοδείκτη cd79a των Β-λεμφοκυττάρων και του lck των Τ-λεμφοκυττάρων. Τα Pkhd1l1 knockout ποντίκια δεν εμφανίζουν κανένα σύμπτωμα στις κινούμενες βλεφαρίδες.</dc:description><dc:description>This study examines the molecular and cellular mechanism of function of the Pkhd1l1 gene in vertebrates. Foxj1a, a master transcription factor of motile ciliogenesis, upregulates both pkhd1l1 zebrafish genes (pkhd1l1α and pkhd1l1β).  Structurally, zebrafish Pkhd1l1 proteins resemble the human PKHD1 protein, which causes autosomal recessive polycystic kidney disease. We hypothesized that both pkhd1l1 zebrafish genes are involved in motile ciliogenesis. To examine our hypothesis, we generated double pkhd1l1 zebrafish knockouts. We also examined Pkhd1l1 knockout mice for motile cilia defects. Double pkhd1l1 zebrafish knockouts show otolith defects 24 hpf, but they do not show any other motile-cilia defects. Double pkhd1l1 zebrafish knockouts show upregulation of the B-cell marker cd79a and the T-cell marker lck.
