The aim of this thesis is the development and application of LC-MS analytical methods in the determination of small endogenous and exogenous molecules (<1000 Da) in food and biological matrices. Throughout its scope, innovative analytical methodologies (e.g. quantification of cyclic oligomers in blood and lactic acid by the inverted curve method), and their applications are described in order to investigate human health. In addition to the targeted methods, non-targeted methods of metabolomic analysis are applied to address biological questions holistically, such as the effect of exercise on the body. Furthermore, a performance comparison of five different chromatographic columns to evaluate their sufficiency of analysis for a large analyte library is described.
Στόχος της παρούσας διατριβής είναι η ανάπτυξη και η εφαρμογή αναλυτικών μεθόδων LC-MS στον προσδιορισμό μικρών ενδογενών και εξωγενών μορίων (<1000 Da) σε τρόφιμα και βιολογικά υποστρώματα. Σε όλη την έκτασή της περιγράφονται καινοτόμες αναλυτικές μεθοδολογίες που έλειπαν μέχρι πρότινος από την παγκόσμια βιβλιογραφία (π.χ. ποσοτικός προσδιορισμός των κυκλικών ολιγομερών στο αίμα και του γαλακτικού οξέος με τη μέθοδο ανεστραμμένης καμπύλης), και οι εφαρμογές τους με σκοπό τη διερεύνηση σύγχρονων ερωτημάτων που αφορούν την ανθρώπινη υγεία. Εκτός από τις στοχευμένες μεθόδους, γίνεται και εφαρμογή μη στοχευμένων μεθόδων μεταβολομικής ανάλυσης για την ολιστική προσέγγιση βιολογικών ερωτημάτων, όπως η επίδραση της άσκησης στον οργανισμό.
