Η ερευνητική αυτή εργασία περιλαμβάνει σειρά συνθετικών προσεγγίσεων και την ανάπτυξη της σχετικής συνθετικής μεθοδολογίας για την παρασκευή σειράς συνθετικών αναλόγων του κοστικού οξέος, μιας ένωσης που απομονώθηκε από το φυτό Dittrichia viscosa (κοιν. Ακονιζά), η οποία θανατώνει το παρασιτικό άκαρι Varroa destructor της μέλισσας χωρίς να βλάπτει την ίδια.
Οι συνθετικές προσεγγίσεις που ακολουθήθηκαν περιλαμβάνουν την προσθήκη επιπλέον καρβοξυλικών ομάδων στον ανθρακικό σκελετό του κοστικού οξέος όπως επίσης και την απλοποίηση του βασικού δικυκλικού συστήματος της ένωσης.
Συγκεκριμένα, σε μια αρχική συνθετική προσέγγιση χρησιμοποιήθηκε σαν αρχικό αντιδρών η εμπορικά διαθέσιμη ένωση 1,7-διυδροξυναφθαλένιο (23) και μέσω μιας σειράς αντιδράσεων έλαβε χώρα η σύνθεση της ένωσης-στόχου 2-(8-μεθόξυ-1,2,3,4-τετράϋδρο-2-ναφθαλενυλο) μαλονικού διμεθυλεστέρα (dimethyl-2-(8-methoxy-1,2,3,4-tetrahydro-2-naphthalenyl)malonate.
Στη συνέχεια, πραγματοποιήθηκε προσπάθεια διαφοροποίησης του σκελετού του κοστικού οξέος ως προς το μεθύλιο που βρίσκεται στο σημείο σύντηξης του δικυκλικού συστήματος. Για το σκοπό αυτό χρησιμοποιήθηκε η εμπορικά διαθέσιμη ένωση κυκλοεξανόνη (60a), όμως από τα πρώτα στάδια της συνθετικής πορείας παρουσιάστηκαν δυσκολίες και το επιθυμητό προϊόν δεν σχηματίστηκε.
Η τελευταία συνθετική προσέγγιση περιλαμβάνει τη χρήση της μεθυλοκυκλοεξανόνης (60b) σαν αρχικό αντιδρών σε μία σειρά εναλλακτικών συνθέσεων με ένωση-στόχο τον 2-(4α-μεθυλο-8-μεθυλενο-2,3,4,4α,5,6,7,8-οκταϋδρο-2-ναφθαλενυλο) μαλονικό διμεθυλεστέρα (dimethyl 2–(4a-methyl-8-methylene-2,3,4,4a,5,6,7,8-octahydro-2-napthalenyl)malonate).

This research project includes a number of synthetic approaches and the development of relevant synthetic methodology for the preparation of a series of synthetic analogs of Costic Acid, a compound isolated from the plant Dittrichia viscosa (Greek: Aconiza), which kills the parasitic mite of honey bee Varroa destructor without damaging the insect .
The synthetic approaches followed, include the introduction of additional carboxylic moieties on the carbon backbone of Costic Acid as well as “structural simplifications” on the bicyclic carbon framework.
Specifically, one of the synthetic approaches used as starting reactant, the commercially available compound of 1,7-dihydroxynaphthalene and through a series of reactions the synthesis of the target compound dimethyl-2-(8-methoxy-1,2,3,4-tetrahydro-2-naphthalenyl) malonate was achieved.
Then, an attempt was made to modify the carbon framework of Costic Acid regarding the presence of the methyl group located at the fusion point of the bicyclic system. To this purpose, commercially available compound cyclohexanone (60a) was used, but, even in the early stages of the synthesis this approach was found to be problematic and the desired products were not formed.
The last synthetic approach involved the use of methylcyclohexanone (60b) as a starting material in a series of approaches to the target compound, namely, dimethyl 2- (4a-methyl-8-methylene-2,3,4,4a, 5,6,7,8-octahydro-2-napthalenyl) malonate.
