Με τον όρο «κυτταρική κλασματοποίηση» εννοούμε τον διαχωρισμό των δομικών συστατικών του κυττάρου με βάση το μέγεθος και την πυκνότητά τους. Στόχος της άσκησης 5 είναι η εκμάθηση των μεθόδων κυτταρικής κλασματοποίησης και η αναγνώριση, απομόνωση, χρώση και μικροσκοπική παρατήρηση διαφόρων υποκυτταρικών οργανιδίων. Στο εισαγωγικό μέρος της άσκησης περιγράφονται οι μέθοδοι ομογενοποίησης και κυτταρικής κλασματοποίησης (φυγοκέντρηση με διαβάθμιση πυκνότητας, διαφορική φυγοκέντρηση, υπερφυγοκέντρηση κ.λπ.), ενώ στο πρακτικό μέρος περιγράφεται και εξηγείται η διαδικασία απομόνωσης πυρήνων και μιτοχονδρίων από ηπατικό ιστό. Επιπλέον, ο φοιτητής μελετά τη μορφολογία των βασικών υποκυτταρικών οργανιδίων στο ζωικό και φυτικό κύτταρο και εκπαιδεύεται στην αναγνώρισή τους με τη βοήθεια εικόνων και μόνιμων αντικειμενοφόρων πλακών μικροσκοπίου ζωικών και φυτικών ιστών. By 'cell fractionation' we mean the separation of the cell's structural components based on their size and density. The aim of Exercise 5 is to learn the methods of cell fractionation and to identify, isolate, stain and microscopically observe various subcellular organelles. The introductory part of the exercise describes the methods of homogenization and cell fractionation (density gradient centrifugation, differential centrifugation, ultracentrifugation, etc.), while the practical part describes and explains the procedure for isolating nuclei and mitochondria from liver tissue. In addition, the student studies the morphology of the basic subcellular organelles in the animal and plant cell and is trained in their identification with the aid of images and permanent microscope slides of animal and plant tissues.
