The aim of this study was the cytogeneting monitoring of chromosomal fragility with stomach cancer. The cytogenetic methodology used in our study, was the Sister Chromatid Exchanges technique. Fluorescence plus Giemsa (FPG) method was used in order to dye chromosomes. FPG method is a developed version of SCEs which helps to avoid using radioisotopes. Proliferation Rate Index (PRI) and Mitotic Index (MI) were also counted. MMC, a chemical antibiotic, which is an original alkyliotic agent, was used in our vitro experiments. MMC is widely in the fight against cancer and it is a genotoxic and cytotoxic agent which induces SCEs and that’s why it was used in our experiments as a positive control. Our results shown, that after MMC addition in human lymphocytes cultures in our in vitro experiments, a large increase in SCEs levels was produced and specially in cells from gastric cancer patients. That shows that cancer patients chromosomes of are very fragile. In conclusion, it appears that the increased SCE levels in peripheral blood lymphocyte chromosomes of gastric cancer patients often results in chromosomal fragility and are likely to be significant signs/findings for early prognosis, selection of appropriate treatment and perhaps discovery of new types of better therapy of gastric cancer.
