The present thesis focuses on the development of advanced analytical methods for the determination of organic pollutants in various matrices (surface-drinking water, honey). Target pollutants, i.e., polychlorinated biphenyls (PCBs), halogenated hydrocarbons, cyanotoxins and polycyclic aromatic hydrocarbons (PAHs), were chosen due to their high toxicity, their widespread occurrence and the difficulty in their determination. In this frame, a new method for the determination of polychlorinated biphenyls in water samples has been developed. Analysis was carried out using gas chromatography with electron capture detector (GC/ECD) after extraction and preconcentration of target analytes by solid phase microextraction (SPME) with a 100μm film thickness polydimethylsiloxane fiber. The target compounds studied, known as “PCB markers”, included one trichloro, one tetrachloro, two pentachloro, two hexachloro and one heptachloro biphenyls, named according to IUPAC PCB 28, PCB 52, PCB 101, PCB118, PCB 138, PCB 153 and PCB 180, respectively. Several parameters affecting the extension of the adsorption process (i.e., immersion versus head space SPME extraction efficiency, addition of salt, adsorption time, extraction temperature, stability of the fiber used) were studied. The response of each analyte was determined in the presence and absence of other congeners at different levels. Head space -solid phase microextraction (HS-SPΜE) showed better sensitivity compared to immersion in the aqueous phase, without carryover between the samples. The above method also showed good linearity over a wide concentration range from 0,001μg l-1 to 10 μg l-1 level for the majority of the target analytes, while correlation coefficients (r2) ranged among 0,9735 and 0,9942. The detection limits were in the order of nanogram per liter level (1 to 5 ngl-1) with relative standard deviation (RSD) values below 13% within the above mentioned concentration range. The method was applied for the determination of potable water samples of the area of Athens. Target analytes were not detected or gave values below the limit of quantification of the proposed method. The proposed method proved to be simple, fast sensitive and applicable for the analysis of PCBs in water samples.A new method for the simultaneous determination of halogenated hydrocarbons (1,4-dichlorobenzene (p-DCB), naphthalene and 1,2-dibromoethane (1,2-DBE)) residues in honey has also been developed. Analysis was carried out using gas chromatography–mass spectrometry (GC/MS) in selected ion monitoring mode (SIM), after extraction and preconcentration of target analytes by headspace solid-phase microextraction (HS-SPME), with a 100μm film thickness polydimethylsiloxane (PDMS) fiber. Several parameters affecting the extension of the adsorption process (i.e., addition of salt, extraction time, extraction temperature) were studied. The optimal conditions for the determination of these analytes were established. The proposed HS-SPME method showed good sensitivity, without carryover between the samples. Linearity was studied from 5 to 2500 μgkg−1 for p-DCB, 0.5 to 500 μgkg−1 for naphthalene and 5 to 500 μgkg−1 honey for 1,2-DBE with correlation coefficients (r2) ranging from 0.9901 to 0.9999. Precision was assessed and both intra and inter-day R.S.D.s (%) were below 6.3%. The detection limits were found to be 1, 0.1 and 2 μgkg−1 honey for p-DCB, naphthalene and 1,2-DBE, respectively. The percentage recoveries that were evaluated with the proposed HS-SPME method and the standard addition calibration technique gave values among 72.8 and 104.3% for measurements in samples spiked with one target analyte or mixtures of the three. This method has been applied for the analysis of unknown honey samples. The results showed an excellent applicability of the proposed method for the determination of the target compounds in honey samples.Blooms of cyanobacteria (blue-green algae) occur world-wide in fresh and brackish waters. These blooms pose an environmental hazard because of the release, by some of these species, of water soluble toxic compounds, called cyanotoxins. A part of this thesis has been the development of a new method for the determination of 5 cyanotoxins (MC-RR, -LR, -YR, -LA and nodularin) in surface and drinking water, using liquid chromatography hyphenated with electrospray ionization mass spectrometry (LC-ESI-MS/MS). The extraction and preconcentration of cyanotoxins from spiked samples was carried out with solid phase extraction using Oasis HLB cartridges. The mean recoveries that were evaluated at two concentration levels (0.1 and 1 μgl-1) gave values among 69 and 113 % for all target analytes, while the detection limits ranged among 0.002 and 0.017 μgl-1. The detection, confirmation and quantitative determination were achieved with mass spectrometry-triple stage quadrupole detector in ESI mode, after optimising several parameters. All target compouds were confirmed in SRM mode (one parent ion and two daughter ions for each compound). Fragmentation patterns of the target compounds were explained by proposed mechanisms. The application of the proposed method was carried out in monitoring surface (Marathonas Lake, a reservoir of Athens drinking water) and drinking water of Athens. Cyanotoxins were not detected in drinking water samples but microcystins LR and RR were detected in surface water samples of Marathonas lake at concentrations lower than the limit set in drinking water by WHO (1 μgl-1 for MC-LR). The results of this study constitute the first report on the presence of microcystins in Marathonas Lake. A novel method for the determination of EU 8 priority pollutants polycyclic aromatic hydrocarbons (PAHs), in lake and drinking water, has been developed and validated using liquid chromatography/dopant assisted atmospheric-pressure photoionization/tandem mass spectrometry (LC-APPI-MS/MS). Clean up procedure for lake and drinking water was performed with (SPE) on C18 cartridges. All measurements were carried out in a LC-MS/MS system and the target compounds were quantified by use of 3 deuterated PAHs. Various dopants and dopant mixtures were tested for their effectiveness and the most suitable one for the majority of target compounds was anisole. The proposed LC-APPI-MS/MS method was comprehensively validated taking into account, specificity, linearity, accuracy and precision. Τhe linearity of the method was studied from 1 to 100 μgl-1 for Anthracene, Benzo(b)fluoranthene, Benzo(k)fluoranthene, Benzo(a)pyrene, Benzo(g,h,i)perylene and Indeno(1,2,3,c,d)pyrene and from 10 to 1000 μgl-1 for Naphthalene and Fluoranthene, with correlation coeffcients (r2) ranging from 0.991 to 0.9990. Precision was assessed and both intra and inter-day R.S.D.s (%) were below 19.5 % for all target analytes in surface and drinking water. The mean recoveries, that were evaluated at two concentration levels (0.01 and 0.1 μgl-1/PAH with the exception of Naphthalene and Fluoranthene which were evaluated at the concentration levels 0.1 and 1 μgl-1), were among 78 and 119 % for all target analytes. The proposed analytical method achieves very low limits of detection, in surface waters (<0.005 μgl-1) and drinking waters (<0.0005 μgl-1). To the best of our knowledge this is the first report on PAHs determination in water samples with LC-APPI-MS/MS. Moreover the proposed method is the only LC-MS/MS method which is in compliance with the recent directive 2008/105/EC, providing higher sensitivity and specificity than did earlier published methods. The method was successfully applied in lake and drinking water samples of the area of Athens.
Στην παρούσα διατριβή αναπτύχθηκαν προηγμένες αναλυτικές μέθοδοι για τον προσδιορισμό οργανικών ρύπων σε διαφορετικά υποστρώματα (επιφανειακά και πόσιμα ύδατα, μέλι). Οι οργανικοί ρύποι που επιλέχθηκαν περιλαμβάνουν τα πολυχλωριωμένα διφαινύλια, αλογονομένους υδρογονάνθρακες, κυανοτοξίνες καθώς και τους πολυκυκλικούς αρωματικούς υδρογονάνθρακες. Οι ρύποι αυτοί παρουσιάζουν αυξημένο ερευνητικό ενδιαφέρον στην Ελλάδα αλλά και διεθνώς, λόγω της υψηλής τοξικότητάς τους, της εκτεταμένης εμφάνισής τους καθώς και της μεγάλης δυσκολίας στον προσδιορισμό τους.Ειδικότερα, αναπτύχθηκε μέθοδος προσδιορισμού επτά πολυχλωριωμένων διφαινυλίων (Polychlorinated Biphenyls, PCBs) σε δείγματα πόσιμων και επιφανειακών υδάτων με χρήση της τεχνικής μικροεκχύλισης στερεάς φάσης (Head Space Solid Phase Microextraction, HS-SPΜE). Για το σκοπό αυτό επιλέχθηκαν συγκεκριμένα ισομερή πολυχλωριωμένων διφαινυλίων, τα οποία αποτελούν δείκτες περιβαλλοντικής ρύπανσης (“PCB markers”). Πραγματοποιήθηκε βελτιστοποίηση των πιο σημαντικών πειραματικών παραμέτρων όπως, επιλογή ίνας, χρόνου και θερμοκρασίας εκρόφησης των αναλυτών, επίδραση του χρόνου ανάδευσης και εκχύλισης, η επίδραση μίγματος PCBs στην εκχύλιση κάθε αναλύτη ξεχωριστά. Ακολούθησε επικύρωση της μεθόδου σύμφωνα με τους ισχύοντες κανόνες επικύρωσης αναλυτικών μεθόδων όπως αυτοί περιγράφονται με διεθνή πρότυπα. Η εφαρμοσιμότητα της μεθόδου σε πραγματικά δείγματα υδάτων εκτιμήθηκε με την ανάλυση δειγμάτων πόσιμου νερού από το δίκτυο ύδρευσης της ΕΥΔΑΠ στην Αθήνα. Τα αποτελέσματα έδειξαν ότι δεν ανιχνεύθηκαν μετρήσιμες συγκεντρώσεις PCBs ή οι συγκεντρώσεις ήταν μικρότερες του ορίου ποσοτικοποίησης της μεθόδου. Παράλληλα, διαπιστώθηκε ότι πρόκειται για μία εξαιρετικά απλή, γρήγορη, εφαρμόσιμη και ευαίσθητη μέθοδο. Αλογονομένοι υδρογονάνθρακες όπως το παραδιχλωροβενζόλιο (p-dichlorobenzene, p-DCB), το 1,2 διβρωμοαιθάνιο (1,2-Dibromoethane, 1,2-DBE) και το ναφθαλένιο (Naphthalene, Naphth) προσδιορίσθηκαν ταυτόχρονα σε δείγματα μελιού. Οι ενώσεις αυτές είναι βλαβερές ή έντονα τοξικές για τον άνθρωπο, εμφανίζονται στο μέλι λόγω της χρήσης τους ως χημικά για την αντιμετώπιση του σκώρου των κερύθρων και επομένως η ανίχνευσή τους σε ίχνη παρουσιάζει σημαντικό ερευνητικό ενδιαφέρον. Για το σκοπό αυτό αναπτύχθηκε και παρουσιάσθηκε στην διεθνή βιβλιογραφία για πρώτη φορά, σύγχρονη αναλυτική μέθοδος για τον ταυτόχρονο προσδιορισμό των παραπάνω ρύπων σε δείγματα μελιού. Η μέθοδος βασίζεται στο συνδυασμό των τεχνικών HS-SPME και αέριας χρωματογραφίας-φασματομετρίας μαζών (GC-MS). Η βελτιστοποίηση της μεθόδου έγινε για το σύνολο των πειραματικών μεγεθών που επηρεάζουν την ανάκτηση των τριών αναλυτών με την τεχνική HS-SPME και ακολούθησε η επικύρωσή της με έλεγχο της γραμμικότητας, επαναληψιμότητας, αναπαραγωγισημότητας, των ορίων ανίχνευσης καθώς και της ανάκτησης των τριών αναλυτών σε διαφορετικά επίπεδα συγκέντρωσης. Η μέθοδος εφαρμόσθηκε με επιτυχία σε πραγματικά δείγματα μελιού, εμφανίζοντας απλότητα και ταχύτητα στην εφαρμογή της, καθώς και υψηλή ευαισθησία στην ανίχνευση των συγκεκριμένων αναλυτών. Τα χαρακτηριστικά αυτά καθιστούν την μέθοδο εφαρμόσιμη σε αναλύσεις ρουτίνας και κατάλληλη για την ανάλυση μεγάλου αριθμού δειγμάτων.Οι κυανοτοξίνες αποτελούν οργανικούς ρύπους βιολογικής προέλευσης με υψηλή τοξικότητα και εμφανίζονται τελευταία με αυξημένη συχνότητα σε συστήματα επιφανειακών και παράκτιων υδάτων σε όλο τον κόσμο. Στα πλαίσια της διατριβής, αναπτύχθηκε μια αξιόπιστη μέθοδος υγρής χρωματογραφίας-φασματομετρίας μαζών, τεχνολογίας τριπλού τετραπόλου με ηλεκτροψεκασμό (Electron Spray Ionization, Liquid Chromatography – Tandem Mass Spectrometry, ESI-LC-MS/MS) για τον προσδιορισμό πέντε κυανοτοξινών (μικροκυστίνη- LR, RR, YR, LA και Νοντουλαρίνη) σε πόσιμα και επιφανειακά ύδατα. Παράλληλα, διεξήχθη έρευνα για την παρουσία κυανοτοξινών στα επιφανειακά ύδατα και στο πόσιμο νερό της Αθήνας. Η ανίχνευση και ο ποσοτικός προσδιορισμός των πέντε κυανοτοξινών, με βάση το σύστημα MS/MS, επιτεύχθη μετά από τη βελτιστοποίηση μιας σειράς παραμέτρων που είχαν ως στόχο την μεγιστοποίηση του καταγραφόμενου σήματος. Η ταυτοποίηση όλων των κυανοτοξινών που αναλύθηκαν πραγματοποιήθηκε με την παρακολούθηση δύο θυγατρικών ιόντων για κάθε μοριακή μάζα, ενώ προτάθηκαν και οι μηχανισμοί δημιουργίας των θυγατρικών ιόντων. Η εκχύλιση και προσυγκέντρωση των κυανοτοξινών σε εμβολιασμένα δείγματα νερών πραγματοποιήθηκαν με χρήση της τεχνικής της εκχύλισης στερεάς φάσης (Solid Phase Εxtraction, SPE). Tα ποσοστά ανάκτησης αξιολογήθηκαν σε δύο επίπεδα συγκέντρωσης, ενώ παράλληλα προσδιορίστηκαν εξαιρετικά χαμηλά όρια ανίχνευσης. Η μέθοδος εφαρμόσθηκε για τον συστηματικό έλεγχο δειγμάτων επιφανειακού νερού από τη λίμνη του Μαραθώνα και δειγμάτων πόσιμου νερού από το δίκτυο της ΕΥΔΑΠ στην Αθήνα. Καμία κυανοτοξίνη δεν ανιχνεύθηκε στα δείγματα πόσιμου νερού που εξετάστηκαν, ανιχνεύθηκαν, όμως, μικροκυστίνες (μικροκυστίνη LR και μικροκυστίνη RR) σε δείγματα επιφανειακού νερού από την λίμνη του Μαραθώνα. Οι συγκεντρώσεις που μετρήθηκαν ήταν πολύ χαμηλότερες του ορίου που έχει προταθεί από τον Παγκόσμιο Οργανισμό Υγείας (World Health Organization, WHO) (1 μg/L για την μικροκυστίνη LR). Θα πρέπει να σημειωθεί ότι τα αποτελέσματα της παρούσας μελέτης αποτελούν την πρώτη αναφορά για την ύπαρξη μικροκυστινών στο νερό της λίμνης του Μαραθώνα. Η τελευταία κατηγορία ρύπων που μελετήθηκε στα πλαίσια της διατριβής είναι οι πολυκυκλικοί αρωματικοί υδρογονάνθρακες (ΠΑΥς) (Polycyclic Aromatic Hydrocarbons, PAHs). Οι ενώσεις αυτές προέρχονται τόσο από φυσικές πηγές όσο και από ανθρωπογενείς δραστηριότητες, χαρακτηρίζονται από μεταλλαξιογόνες και καρκινογόνες ιδιότητες και σύμφωνα με την Ευρωπαϊκή και Εθνική νομοθεσία αποτελούν ουσίες προτεραιότητας και η παρουσία τους στα νερά θα πρέπει να ελέγχεται. Συγκεκριμένα, αναπτύχθηκε και επικυρώθηκε μέθοδος προσδιορισμού οκτώ πολυκυκλικών αρωματικών υδρογονανθράκων, με την τεχνική της υγρής χρωματογραφίας - φασματομετρίας μαζών, τεχνολογίας τριπλού τετραπόλου σε συνδυασμό με φωτοϊοντισμό ατμοσφαιρικής πίεσης (LC-APPI-MS/MS). Η εκχύλιση και προσυγκέντρωση των ΠΑΥς σε εμβολιασμένα δείγματα νερών πραγματοποιήθηκε με την τεχνική της εκχύλισης στερεάς φάσης. Για την ποσοτικοποίηση των αναλυτών χρησιμοποιήθηκαν δευτεριωμένα πρότυπα ΠΑΥς. Η βελτιστοποίηση των συνθηκών φωτοϊοντισμού επιτεύχθηκε με τη χρήση ενώσεων που ενίσχυαν τον ιοντισμό των ΠΑΥς (dopants). Η μέθοδος που αναπτύχθηκε, επικυρώθηκε σύμφωνα με διεθνή πρότυπα μετά από διερεύνηση της γραμμικότητας, της ακρίβειας, της εκλεκτικότητας και της επαναληψιμότητας. Τα όρια ανίχνευσης των ΠΑΥς που επιτεύχθηκαν ήταν εξαιρετικά χαμηλά και η μέθοδος ικανοποιεί απόλυτα τις απαιτήσεις πρόσφατης Ευρωπαϊκής Οδηγίας (2008/105/ΕΚ) ως προς τα νέα χαμηλότερα όρια ανίχνευσης και ποσοτικοποίησης των ΠΑΥς σε επιφανειακά νερά. Η εφαρμοσιμότητα της μεθόδου πραγματοποιήθηκε σε πραγματικά δείγματα υδάτων (πόσιμα και επιφανειακά).
