Although the behavior of the Mediterranean fruit fly Ceratitis capitata(Wiedemann) (Diptera: Tephritidae) has been studied in great extent, theeffect of age on medfly behavior remains poorly understood. This informationis missing regarding other Tephritids and insect – model systems, in general.The aim of the current doctorate dissertation was to elucidate the effect of age(with emphasis to extreme ages) and of other factors, such as diet andmating, on the behavior of adult medflies. Experiments were conducted, underconstant laboratory conditions (25 ± 1 °C), in the Laboratory of Entomologyand Agricultural Zoology, University of Thessaly, Greece during 2008 – 2013and are deployed in Chapters 3 to 8. Flies used in the experiments wereobtained from field infested fruits (collected from the area of Volos, Greece)and were reared under laboratory conditions for 1 – 3 generations (F1 – F3).General introductory information regarding insect behavior, theories of aging,age-related behavioral alterations in insects and the medfly are elaborated inChapter 1. The utilization and application of materials and methods during theexperiments are presented in Chapter 2. Conclusions drawn by this thesis,theoretical and practical applications and future research opportunities arediscussed in Chapter 9.In Chapter 3 we studied the effect of diet (sugar, sugar plus yeasthydrolysate) on the daily activity of males and females during their life course.Following a 2-hours daily observation (10:00 – 12:00), we recorded thefrequency of expression of 7 common behaviors (feeding, drinking, walking,resting, sexual signaling of males, oviposition attempts of females and supinebehavior). The lifespan of all adult medflies was also recorded. Resting andsupine behavior were grouped as “inactivity” whilst the rest of the behaviorswere grouped as “activity”. Adult medflies fed the full diet (sugar plus yeasthydrolysate) lived significantly longer than those fed only sugar. Males fed thefull diet were more active than males fed only sugar during their entirelifespan. Nevertheless, females fed the full diet were less active than femalesfed only sugar. Males were more active than females regardless of the dietand the age. Adult activity decreased in older ages regardless of the diet.Despite the fact that protein-fed males were more active than protein-deprived18males, in total, their daily frequency of walking was significantly lower. Thedaily frequency of walking for protein-deprived females was higher than that offemales fed the full diet. Females exhibited a higher walking frequency thanmales regardless of the age and the food regime.In Chapter 4, we studied the effect of diet (sugar, sugar plus yeasthydrolysate) and age on the daily rates of sexual signaling of male medflies.Males fed the full diet reached sooner the reproductive maturity and exhibitedhigher rates of sexual signaling than males fed only sugar during their entirelifespan, including extreme ages. Aging reduced the rates of sexual signalingregardless of the diet regime. A clear age-related pattern of sexual signalingwas exhibited by males fed the full diet, characterized by a short period ofnon-signaling activity until attainment of sexual maturity, followed by a longperiod of frequent sexual signaling and concluded by a short period of pausein signaling activity, prior to death for the long-lived males. High rates ofsexual signaling were positively correlated with longevity in protein-fed males.However, no clear pattern of sexual signaling in relation to age and nocorrelation (positive or negative) between signaling activity and longevity wererecorded for the protein-deprived males.In Chapter 5, we examined the relationship between the period ofunhealthy life and longevity in the medfly by using the first sign of supinebehavior (upside down, immobile) in medflies as an indication of their poorhealth and by altering cohort longevity through dietary manipulations. Forlonger lived medflies it was more likely to observe the supine behavior whileshorter lived flies would more likely die before exhibiting the supine behavior.Males had similar total life expectancy as females but a longer healthy lifeexpectancy. The total number of healthy days, spent in the pre-supine period,and the amount of healthy life span as a fraction of the total lifespan variedwith both sex and diet. Despite the large difference in longevity between bothmale and female medflies when reared on sugar-only diet versus on full diet,the differences in the fraction of their lifespans in the healthy state wererelatively modest (64 vs 61% in females and 77 vs 72% in males). This findingthat there is no significant change in the proportion of the life course in theunhealthy state is consistent with the “dynamic equilibrium” model of healthyaging.19In Chapter 6, we explored the effect of diet manipulation [diet switchfrom sugar only to sugar plus yeast hydrolysate (protein) and vice versa] onthe sexual signaling and the supine behavior of male medflies. Following a 2-hours daily observation (10:00 – 12:00) we recorded the sexual signaling andthe supine behavior of each male throughout the life course. Diet switch wasapplied at the age of 15 days that defines the attainment of sexual maturity inwild male medflies. Protein deprivation after day 15th decreased significantlymale survival while protein availability at the same age increased survival incomparison to males that had continuous access to full and sugar only diet,respectively. The life expectancy of males with a constant access to full dietwas similar to that of males that obtained access to full diet after sexualmaturity. Moreover, the life expectancy of males with constant access to asugar only diet was similar to that of males that were deprived of protein afterattaining sexual maturity. Diet significantly affected both sexual signaling andsupine behavior. The frequency of sexual signaling of males that had constantaccess to full diet was higher throughout their entire lifespan compared tomales that fed a sugar only diet. Males fed only sugar until the age of 15 daysand then switched to a full diet performed similar sexual signaling rates withmales fed continuously a full diet. On the contrary, males fed a full diet untilthe age of 15 days and then switched to a sugar only diet reduced significantlythe frequency of sexual signaling. Males that gained access to a full diet afterattaining sexual maturity increased rapidly their sexual signaling, while malesthat had access to a full diet only until sexual maturity maintained their sexualsignaling in relatively high levels up to day 50. Long-lived males exhibitedsignificantly higher rates of sexual signaling when the diet switched fromsugar only to sugar and protein but not when it switched from sugar andprotein to sugar only. Males fed continuously a full diet exhibited supinebehavior more frequently and for longer periods. Supine behavior increased inolder ages in all dietary protocols.In Chapter 7, we tested the effect of age on the mating competitiveness(mating success against younger rivals) and the mating performance (matingsuccess in conditions lacking competition) of males. The mating probability ofolder males decreased significantly when competing with younger males.Aging gradually reduced the mating performance but older males were still20accepted as mating partners in conditions lacking competition. Therefore,older males were capable of performing the complete repertoire of sexualperformance but failed to be chosen by females in the presence of youngrivals. Older males achieved shorter copulations than younger ones, andfemale readiness to mate was negatively affected by male age. Older andyounger males transferred similar amount of spermatozoids to femalespermathecae. Females stored spermatozoids asymmetrically in the twospermathecae regardless the age of their mating partner. Aging positivelyaffected the amount of spermatozoids in testes of both mated and non-matedmales. No significant differences were observed on the amount ofspermatozoids between mated and non-mated males.Finally, in Chapter 8, we studied the effects of protein deprivation andage of mating (young – 15 days old and old – 40 days old) on female fitnesstraits (survival and fecundity). We recorded the lifespan, fecundity andcopulation characteristics (latency to mate and copulation duration) of femalesmated at young or at old age and fed a full or a protein deprived diet. Thefindings suggest that condition (nutritional status and age) defined the positiveor negative output of mating in female medflies. Old females were morereceptive and less selective in mating than young females regardless of thefood regime. Mating significantly increased the longevity of protein-deprivedfemales. On the contrary, mating boosted the egg production and reduced thelongevity of protein-fed females. Mortality responses (negative or positive) tomating were expressed after a long lag phase.The results of the current thesis contribute to the “behavioral mapping”of the Mediterranean fruit fly, an important model organism for aging researchand a major pest of agricultural commodities. The effects of three vitalconditions (age, age of mating, adult diet) on selected fitness traits of bothmales and females were also explored. The results demonstrate thatimportant behaviors of adult medflies, such as copulation, sexual signaling,oviposition and supine behavior, may affect several fitness components andinduce a cost on survival of both sexes, depending on the condition. Thecontribution of these findings towards understanding the behavior of themedfly, as well as their implications for the research of aging are beingdiscussed and possibilities for future research are being proposed.
Μελετήθηκε σε σταθερές συνθήκες στο εργαστήριο (25 °C) ησυμπεριφορά ενηλίκων της μύγας της Μεσογείου, Ceratitis capitata(Wiedemann) (Diptera: Tephritidae) με έμφαση στις μεγαλύτερες ηλικίες (40 –70 ημερών). Χρησιμοποιήθηκαν έντομα που προήλθαν απόπροσβεβλημένους καρπούς που συλλέχθηκαν στην περιοχή των Λεχωνίωνκαι της Αγχιάλου του νομού Μαγνησίας και εκτράφηκαν για 1 έως 3 γενεές (F1– F3) στο εργαστήριο.Στην πρώτη ενότητα πειραμάτων (Κεφάλαιο 3) μελετήθηκε σεεργαστηριακές συνθήκες η μεταβολή της συμπεριφοράς αρσενικών καιθηλυκών, που τρέφονταν με ζάχαρη ή με ζάχαρη και πρωτεΐνη, καθ’ όλη τηδιάρκεια της ζωής τους. Οι παρατηρήσεις πραγματοποιούνταν καθημερινάαπό τις 10:00 έως τις 12:00 και καταγράφονταν εφτά βασικές συμπεριφορές[διατροφική δραστηριότητα, πρόσληψη νερού, βάδιση, ηρεμία, εκδήλωσησεξουαλικού καλέσματος για τα αρσενικά, προσπάθεια ωοτοκίας για ταθηλυκά και εκδήλωση ύπτιας συμπεριφοράς (supine)]. Επιπλέον,καταγράφηκε η επιβίωση όλων των ενηλίκων. Οι συμπεριφορέςομαδοποιήθηκαν σε δύο κατηγορίες: δραστηριότητα και αδράνεια. Στηνκατηγορία της δραστηριότητας συμπεριλήφθηκαν η βάδιση, η πρόσληψητροφής και νερού, η προσπάθεια ωοτοκίας των θηλυκών και το σεξουαλικόκάλεσμα αρσενικών. Στην κατηγορία της αδράνειας συμπεριλήφθηκαν ηανάπαυση και η εκδήλωση ύπτιας συμπεριφοράς. Η ανάλυση τωναποτελεσμάτων έδειξε ότι αρσενικά και θηλυκά που τρέφονταν με ζάχαρη καιπρωτεΐνη έζησαν περισσότερο από αυτά που τρέφονταν μόνο με ζάχαρη. Τααρσενικά που τρέφονταν με ζάχαρη και πρωτεΐνη ήταν περισσότεροδραστήρια σε κάθε ηλικιακή κλάση σε σχέση με αυτά που τρέφονταν μόνο μεζάχαρη. Ωστόσο, τα θηλυκά που τρέφονταν με ζάχαρη και πρωτεΐνηεκδήλωσαν μικρότερη δραστηριότητα από τα θηλυκά που τρέφονταν μόνο μεζάχαρη. Τα αρσενικά ήταν συνολικά περισσότερο δραστήρια από τα θηλυκάανεξάρτητα από το είδος της τροφής και την ηλικία. Η δραστηριότητα τωνενηλίκων μειωνόταν σε σχέση με την ηλικία, ανεξάρτητα από το είδος τηςτροφής. Η συχνότητα βάδισης των αρσενικών που τρέφονταν με ζάχαρη καιπρωτεΐνη ήταν σημαντικά χαμηλότερη σε σχέση με τα αρσενικά που12στερούνταν πρωτεΐνης, παρά το γεγονός ότι συνολικά εκδήλωναν εντονότερηδραστηριότητα. Τα θηλυκά που τρέφονταν μόνο με ζάχαρη εκδήλωνανυψηλότερη συχνότητα βάδισης, επιβεβαιώνοντας τη συνολικά εντονότερηδραστηριότητά τους σε σχέση με τα θηλυκά που τρέφονταν με ζάχαρη καιπρωτεΐνη. Τα θηλυκά εκδήλωσαν υψηλότερη συχνότητα βάδισης σε σχέση μετα αρσενικά ανεξάρτητα από την ηλικία και την τροφή.Στο Κεφάλαιο 4 μελετήθηκε η επίδραση της τροφής και της ηλικίαςστην εκδήλωση σεξουαλικού καλέσματος των αρσενικών. Αρσενικά πουτρέφονταν με ζάχαρη και πρωτεΐνη ωρίμασαν γρηγορότερα σεξουαλικά καιεκδήλωναν σεξουαλικό κάλεσμα με σημαντικά υψηλότερη συχνότητα σεσχέση με τα αρσενικά που τρέφονταν μόνο με ζάχαρη καθ’ όλη τη διάρκειατης ζωής τους, συμπεριλαμβανομένων και των μεγαλύτερων ηλικιών. Η ηλικίαεπέδρασε αρνητικά στη συχνότητα του σεξουαλικού καλέσματος ανεξάρτητααπό τη διαθεσιμότητα πρωτεΐνης στην τροφή. Τα αρσενικά που τρέφονταν μεζάχαρη και πρωτεΐνη εμφάνισαν ξεκάθαρο μοτίβο εκδήλωσης σεξουαλικούκαλέσματος σε σχέση με την ηλικία, που χαρακτηρίζεται από μια σύντομηπερίοδο αδράνειας έως τη σεξουαλική ωρίμανση την οποία ακολουθεί μιαμεγάλη περίοδος συχνής εκδήλωσης σεξουαλικού καλέσματος καιολοκληρώνεται από μια σύντομη περίοδο αδράνειας έως το θάνατο, σταμακρόβια αρσενικά. Η υψηλή συχνότητα εκδήλωσης σεξουαλικού καλέσματοςεμφάνισε θετική συσχέτιση με την επιβίωση στα αρσενικά που είχανπρόσβαση σε πρωτεΐνη. Τα αρσενικά που τρέφονταν μόνο με ζάχαρη δενεμφάνισαν ξεκάθαρο μοτίβο εκδήλωσης σεξουαλικού καλέσματος σε σχέση μετην ηλικία ούτε και συσχέτιση της μακροβιότητας με τη συχνότητα εκδήλωσήςτου.Η επόμενη ενότητα (Κεφάλαιο 5) επικεντρώθηκε στην επίδραση τηςηλικίας και της τροφής στην εκδήλωση της ύπτιας συμπεριφοράς (supine) τωνενηλίκων της μύγας της Μεσογείου. Η περίοδος εκδήλωσης ύπτιαςσυμπεριφοράς ορίστηκε ως μη υγιής και έγινε προσπάθεια καθορισμού τηςσχέσης της με τη μακροβιότητα και με την υγιή περίοδο ζωής των ενηλίκων.Τα αποτελέσματα έδειξαν ότι τα μακροβιότερα άτομα εκδήλωναν ύπτιασυμπεριφορά ενώ τα βραχύβια συνήθως πέθαιναν πριν την εκδήλωση τηςύπτιας συμπεριφοράς. Αρσενικά και θηλυκά είχαν παραπλήσιο προσδόκιμοζωής αλλά τα αρσενικά εκδήλωσαν μεγαλύτερο προσδόκιμο υγιούς ζωής. Το13φύλο και η τροφή είχαν σημαντική επίδραση στη διάρκεια της υγιούς περιόδουπριν την εκδήλωση της ύπτιας συμπεριφοράς καθώς και στη συνολικήδιάρκεια της υγιούς ζωής. Παρά τις μεγάλες διαφορές στη μακροβιότητα τωνθηλυκών που τρέφονταν μόνο με ζάχαρη σε σχέση με αυτά που τρέφονταν μεζάχαρη και πρωτεΐνη, οι διαφορές στη διάρκεια της υγιούς περιόδου της ζωήςήταν μικρές (64% και 61%, ζάχαρη και ζάχαρη και πρωτεΐνη, αντίστοιχα).Παραπλήσια ήταν τα αποτελέσματα και για τα αρσενικά με τη διάρκεια τηςυγιούς περιόδου της ζωής να αποτελεί το 77% και το 72% της συνολικήςδιάρκειας ζωής για τα αρσενικά που τρέφονταν μόνο με ζάχαρη και με ζάχαρηκαι πρωτεΐνη, αντίστοιχα. Τα αποτελέσματα της μελέτης αυτής συμφωνούν μετο μοντέλο «δυναμικής ισορροπίας» της υγιούς γήρανσης, το οποίουποστηρίζει ότι η διάρκεια της μη υγιούς περιόδου και η συνολική διάρκειαζωής μεταβάλλονται αναλογικά.Στο Κεφάλαιο 6 μελετήθηκε η επίδραση της εναλλαγής της τροφής(μεταφορά από ζάχαρη σε ζάχαρη και πρωτεΐνη και αντίστροφα) στηνεπιβίωση, τη συχνότητα εκδήλωσης του σεξουαλικού καλέσματος και τησυχνότητα εκδήλωσης ύπτιας συμπεριφοράς των αρσενικών. Οιπαρατηρήσεις πραγματοποιούνταν καθημερινά από τις 10:00 έως τις 12:00και η εναλλαγή της τροφής πραγματοποιήθηκε στην ηλικία των 15 ημερών,που ορίζει την ολοκλήρωση της σεξουαλικής ωρίμανσης για τα άγριααρσενικά της μύγας της Μεσογείου. Ο διατροφικός περιορισμός (έλλειψηπρωτεΐνης) μετά το στάδιο της σεξουαλικής ωρίμανσης μείωσε σημαντικά τηνεπιβίωση των αρσενικών ενώ η διαθεσιμότητα πρωτεΐνης μετά την σεξουαλικήωρίμανση αύξησε την επιβίωσή τους. Το προσδόκιμο ζωής ήταν παραπλήσιομεταξύ των αρσενικών που είχαν συνεχώς πρόσβαση σε πρωτεΐνη καιεκείνων που στερούνταν πρωτεϊνούχου τροφής έως την ηλικία των 15ημερών, καθώς και μεταξύ των αρσενικών που είχαν πρόσβαση σε πρωτεΐνημόνο μέχρι τη σεξουαλική τους ωρίμανση και εκείνων που τρέφονταν συνεχώςμόνο με ζάχαρη. Η τροφή επηρέασε σημαντικά τόσο το σεξουαλικό κάλεσμαόσο και τη συχνότητα εκδήλωσης ύπτιας συμπεριφοράς. Αρσενικά πουτρέφονταν συνεχώς με ζάχαρη και πρωτεΐνη εκδήλωναν σεξουαλικό κάλεσμασυχνότερα από τα αρσενικά που τρέφονταν μόνο με ζάχαρη καθ’ όλη τηδιάρκεια ζωής τους. Αρσενικά που τρέφονταν με ζάχαρη έως την ηλικία των15 ημερών και στη συνέχεια με ζάχαρη και πρωτεΐνη εκδήλωναν σεξουαλικό14κάλεσμα σε επίπεδα παραπλήσια με τα αρσενικά που είχαν συνεχώςπρόσβαση σε πρωτεΐνη. Αντίθετα, εκείνα που τρέφονταν με ζάχαρη καιπρωτεΐνη έως την ηλικία των 15 ημερών και στη συνέχεια μόνο με ζάχαρημείωσαν σημαντικά τη συχνότητα εκδήλωσης σεξουαλικού καλέσματος μετάτην εναλλαγή της τροφής. Αρσενικά που απέκτησαν πρόσβαση σε πρωτεΐνημετά τη σεξουαλική ωρίμανση αύξησαν άμεσα το σεξουαλικό κάλεσμα, ενώεκείνα που είχαν πρόσβαση σε πρωτεΐνη μόνο έως τη σεξουαλική ωρίμανσηδιατήρησαν το σεξουαλικό κάλεσμα σε σχετικά υψηλά επίπεδα έως την ηλικίατων 50 ημερών. Η εκδήλωση σεξουαλικού καλέσματος ήταν σημαντικάυψηλότερη στα μακρόβια αρσενικά όταν η εναλλαγή της τροφήςπραγματοποιήθηκε από ζάχαρη σε ζάχαρη και πρωτεΐνη, όχι όμως ότανπραγματοποιήθηκε από ζάχαρη και πρωτεΐνη σε ζάχαρη. Η ύπτιασυμπεριφορά ήταν συχνότερη και εκδηλώθηκε για μεγαλύτερη χρονικήπερίοδο και από μεγαλύτερο ποσοστό αρσενικών που τρέφονταν συνεχώς μεζάχαρη και πρωτεΐνη και εντάθηκε σε προχωρημένες ηλικίες σε όλα ταδιατροφικά πρωτόκολλα.Στο Κεφάλαιο 7, μελετήθηκε η επίδραση της ηλικίας στηνανταγωνιστικότητα σύζευξης (επιτυχία σύζευξης σε συνθήκες ανταγωνισμούμε νεαρά αρσενικά) και στην ικανότητα σύζευξης (επιτυχία σύζευξης σεσυνθήκες έλλειψης ανταγωνισμού) των αρσενικών της μύγας της Μεσογείου.Πραγματοποιήθηκαν δοκιμές σύζευξης με θηλυκά που είχαν τη δυνατότηταεπιλογής ανάμεσα σε νεαρά και σε αρσενικά μεγαλύτερης ηλικίας και δοκιμέςσύζευξης χωρίς επιλογή, με αρσενικά συγκεκριμένης ηλικίας. Η πιθανότητασύζευξης αρσενικών μεγάλης ηλικίας μειώθηκε δραματικά σε συνθήκεςανταγωνισμού με νεαρότερα αρσενικά. Η ικανότητα σύζευξης των αρσενικώνμειώθηκε σταδιακά σε σχέση με την ηλικία αλλά γηραιότερα αρσενικάγίνονταν αποδεκτά για σύζευξη από τα θηλυκά σε ικανοποιητικό βαθμόαπουσία ανταγωνισμού. Γηραιότερα αρσενικά πέτυχαν μικρότερης διάρκειαςσυζεύξεις απ’ ότι τα νεαρά αρσενικά ενώ ο χρόνος έως τη σύζευξη, πουυποδηλώνει την επιλεκτικότητα των θηλυκών για σύζευξη, αυξανόταν σεσχέση με την ηλικία των αρσενικών. Η ποσότητα των σπερματοζωαρίων πουμετέφεραν τα αρσενικά στις σπερμοθήκες των θηλυκών δεν επηρεάστηκε απότην ηλικία των αρσενικών. Τα θηλυκά αποθήκευαν ασύμμετρα το σπέρμα στιςσπερμοθήκες τους ανεξάρτητα από την ηλικία του αρσενικού
